摘  要:目的研究脑深部电刺激(DBS)刺激吗啡成瘾大鼠伏核后,伏核cAMP反应元件蛋白(CREB)含量的变化。方法 40只雄性SD大鼠,随机分组,正常对照组(NS组)、吗啡成瘾组(MA组)、假刺激组(SS组)、刺激组(DBS组)各10只,MA组、SS组和DBS组建立吗啡成瘾大鼠动物模型,SS组和DBS组大鼠双侧伏核植入电极。DBS组大鼠行伏核电刺激,刺激后利用western blotting的方法检测各组大鼠伏核CREB的含量。结果 NS组大鼠伏核CREB含量明显低于MA组,P<0.01;DBS组大鼠伏核CREB含量明显低于MA组,P<0.01;SS组大鼠伏核CREB含量与MA组相比则无明显差异性。结论 DBS电刺激伏核,使吗啡成瘾大鼠伏核CREB含量降低,提示DBS对成瘾大鼠心理依赖的戒断效应可能是通过抑制CREB的表达而起作用。Objective To study the change of the content of cAMP response element protein(CREB)in the nucleus accumbens of morphine addicted rats after the Deep Brain Stimulation(DBS).Methods 40 male SD rats were randomly divided into normal control group(NS group),morphine addiction group(MA group),sham stimulation group(SS group),stimulation group(DBS Group).And each group contained 10 rats.MA group,SS Group and DBS Group were established into the morphine-addicted rat model.Then the SS Group and DBS Group were implanted stimulator,and stimulated DBS Group rats' nucleus accumbens.Western blotting method was used to detect each rat's CREB content of nucleus accumbens.Results the CREB content of NS group nucleus accumbens was significantly lower than the MA group(P0.01);and the CREB content of DBS group nucleus accumbens was significantly lower than the MA group(P0.01);MA group rats' nucleus accumbens CREB levels compared with the SS group had no significant difference.Conclusion DBS stimulated in the rats' nucleus accumbens,and the CREB content decreased in morphine addicted rats,suggesting that DBS may affect psychological dependence on morphine addicted rats by reducing the expression of CREB.
