摘  要:目的探讨Tankyrase 1和自噬与老龄大鼠阴茎勃起功能障碍的相关性。方法不同月龄(2、8、1 6和24月龄)雄性大鼠作为研究对象,神经刺激下阴茎海绵体压力和平均动脉压比值(ICP/MAP)检测用来评价阴茎勃起功能,Masson染色检测大鼠阴茎组织平滑肌细胞和胶原纤维比值(CSMC s/CF),Westernblot检测大鼠阴茎海绵体组织中Tankyrase 1、自噬标记物LC3-Ⅰ/Ⅱ表达和自噬调节蛋白Beclin 1、p70S6K和pho spho-P70S6K(Thr3 89)的表达变化。结果老龄(24月龄)大鼠组ICP/MAP比值和CSMCs/CF比值较其8龄组显著降低(P<0.01)。Western blot检测结果 ,老龄大鼠阴茎海绵体组织中Tankyrase 1、自噬标记物LC3-Ⅰ/Ⅱ及自噬调节蛋白Beclin 1的表达比较8月龄组显著降低(P<0.05)。各月龄组自噬调节蛋白p70S6K表达差异无统计学意义(P>0.05),但是,phospho-p70S6K(Thr389)/p70S6K的比值在老龄组表达显著增加,差异有统计学意义(P<0.05)。结论老龄大鼠勃起功能障碍与阴茎海绵体组织CSMCs含量降低密切相关,其发生机制可能与CSMCs中Tankyrase 1表达降低、与自噬功能减弱有关,有待于进一步研究。Objective To study the relationship of Tankyrase 1 and autophagy on erectile dysfunction of aging rats. Methods Male Wistar rats different ages (2, 8, 16, 24 month-old) were used in the study for erectile function investigation by ICP/MAP test. Morphological changes of corpus cavernosum (CC) were analyzed by masson stain for calculating the ratio of corpus cavernosum smooth muscle cells and collogen fibroblasts (CSMCs/CF). The expres- sions of Tankyrase 1, LC3-I/II, Beclin 1, p70S6K and phospho-pTOS6K (Thr389) in CC were detected by western blot. Results The ratios of ICP/MAP and CSMCs/CF in the 24-month-old rats were significantly lower than that of the 8- month-old rats(P〈0.01). The expressions of Tankyrase l, LC3-I/II and Beclin 1 in CC of 24-month-old rats were lower than that of 8-month-old rats(P〈0.05). No obvious difference in the expression of pTOS6K was found among different age groups (P〉0.05). Whereas, the expression of phospho-pTOS6K(Thr389)/p70S6K in CC of 24-month-old rats was significantly higher than that of 8-month-old rats(P〈0.01). Conclusion The results suggested that ED of aging rats might be associated with the decrease of CSMCS in CC. Down-regulated of Tankyrase 1 and decrease of autophagy function might be the direct reason in senile organic ED, which need further investigation.
