摘 要:酶法生产生物柴油较好的发展前景,但酶的价格过高制约其进一步发展,为降低酶的成本,本试验室从自然界筛选得到产气肠杆菌脂肪酶。以菜籽油为原料,利用自制的固定化酶进行催化生产生物柴油的研究。固定化脂肪酶用量为1 000 U,乙醇:油物质的量比为4:1,乙醇分2次等量加入,5 mL正己烷,180 r/min,35℃,反应48 h,脂肪酸乙酯产率为92.91%。在水含量小于10%时,酯化率大于80%,该反应体系可催化有一定含水量的原料生产生物柴油;油酸含量高有利于生产生物柴油,而芥酸有不利影响。试验表明,筛选得到的产气肠杆菌脂肪酶可较好的用于催化生产生物柴油,该酶的培养周期短、价格低廉,可有效降低酶的成本,促进酶催化法生产生物柴油的产业化发展。Enzyme catalyzed biodiesel has a promising prospect in biodiesel industry,whereas the high cost of enzymes constrains its further development.In order to decrease the cost of lipase,this lab screened a lipase of Enterobacter aerogenes.Using vegetable oil as raw material,the self - made immobilized enzyme was adopted to catalyze the production of bio - diesel in this work.Results show the yield of fatty acid ethyl ester is 92.91%under the fol- lowing conditions:the dosage of diatomite immobilized lipase of Enterobacter aerogenes is 1000 U,adding ethanol twice as acyl receptors and the molar ratio of alcohol:oil is 4-1,n - hexane 5 mL,180 r/min,35℃,and reaction time is 48 h.The esterification rate is over 80%when the raw material contains water below 10%.It is found that high content of oleic acid in raw material leads a better result,while erucic acid is disadvantageous.The lipase of Enterobacter aerogenes has advantages of short culture time and low cost,so it will be a promising alternate lipase for biodiesel production.
