摘  要:目的 建立同时测定复方黄连胶囊中黄连所含小檗碱型生物碱高效液相色谱(HPLC)定量法.方法 采用高效液相色谱法,以Kromasil C18(250 mm×4.6 mm,5 μm)为色谱柱;流动相:乙腈-0.05 mol/L磷酸二氢钾溶液(用磷酸调节pH至3)-乙腈(68∶32);流速:1.0 ml/min;检测波长:268 nm;柱温:40℃.结果 盐酸药根碱、盐酸巴马汀、盐酸小檗碱分别在0.0186~0.155、0.0153~0.1275、0.0102~0.085μg范围内.浓度与峰面积线性关系良好(r=0.9996,0.9998,0.9999).平均回收率分别为99.86%、99.78%、100.46%.结论 高效液相色谱法测定复方双黄连胶囊中小檗碱型生物碱方法简便、快速准确、灵敏度高,重现性好.Objective To establish a method for the determination of protoberberine in compound Huanglian capsule by high performance liquid chromatography ( HPLC). Methods The HPLC system was consisted of Kromasil C18column, 0.05 mol/LKH2PO4(adjust pH to 3 with H3PO4)-acetonitrile(68: 32). Flow rate was 1.0 ml/min and column temperature was 40 ℃. Results The method had good linear relation with 0.0186 ~0.155 μg of tatrorrhizine hydrochloride, 0. 0153 ~ 0. 1275 μg of palmatine hydrochloride and 0. 0102 ~ 0. 085 μg of berberine hydrochloride. The correlation coefficients were 0.9996, 0.9998, 0.9999 respectively. The results of the recovery were 99. 86% (RSD = 1.0% ), 99. 78% ( RSD = 0. 6% ) and 100. 46% ( RSD = 1.6%). Conclusion HPLC is simple, fast, rapid and accurate for the determination of preparation.
