丝胶对糖尿病肾病大鼠肾功能和肾脏TIMP-1表达的影响

摘　　要：目的观察彩色蚕茧提取物-丝胶对糖尿病肾病(DN)大鼠肾功能和金属蛋白酶组织抑制因子-1(TIMP-1)表达的影响。方法 60只雄性SD大鼠随机分为5组(每组12只):正常对照组、DN模型组、丝胶治疗组、二甲双胍组和丝胶预防组。除正常对照组外,其他各组大鼠均采用链脲佐菌素(STZ)制糖尿病动物模型成功后,丝胶治疗组大鼠给予丝胶灌胃(2.4g.kg-1.d-1,35d)、二甲双胍组大鼠给予二甲双胍灌胃(55.33mg.kg-1.d-1,35d),丝胶预防组大鼠于注射STZ前给予同等剂量丝胶灌胃35d。分别检测各组大鼠的血糖、肌酐、尿素氮和24h尿蛋白含量,免疫组化染色观察肾脏TIMP-1蛋白的表达。结果与正常对照组大鼠相比,模型组大鼠的血糖、肌酐、尿素氮、24h尿蛋白含量和肾脏TIMP-1蛋白的表达均明显升高(P<0.01);丝胶治疗组、丝胶预防组和二甲双胍组大鼠的血糖、肌酐、尿素氮、24h尿蛋白含量和肾脏TIMP-1蛋白的表达均明显低于模型组(P<0.01,P<0.05),且丝胶治疗组、丝胶预防组与二甲双胍组比较无明显差别(P>0.05)。结论丝胶可明显改善DN大鼠的肾功能和降低肾脏TIMP-1蛋白的表达,对DN时肾脏基质纤维化和肾小球结构破坏具有良好的保护和预防保护作用,且作用与二甲双胍相当。Objective To observe the effects of color silk cocoon extraction-sericine on renal function and tissue inhibitors of tissue inhibitor of metalloproteinase-1 （TIMP-1） expression in kidney of diabetic nephropathy （DN） rats.Methods 60 male SD rats were randomly divided into 5 groups （n=12）：normal,DN model,sericine treatment,metformin and sericine prevention groups.The rats in model,sericine treatment,metformin and sericine prevention groups were all established DN rats model by intraperitoneally injected streptozotocin （STZ）.Blood glucose ≥16.7 mmol/L was taken to judge if the rats model were successfully established.After the rats model were successfully established：the rats in sericine treatment group were lavaged with sericine （2.4 g·kg-1·d-1,35 d）.The rats in metformin group were lavaged with metformin （55.33 mg·kg-1·d-1,35 d）.The rats in sericine prevention group were lavaged with the same dose sericine for 35 d before injecting STZ.The blood glucose,creatinine,urea nitrogen and 24 hours urine protein of rats in each group were respectively detected.Immunohistochemical staining was used to observe the expression of TIMP-1 in kidney.Results Compared with normal control rats,the blood glucose,creatinine,urea nitrogen,24 hours urine protein and TIMP-1 expression in kidney of model rats increased obviously （P0.01）.The blood glucose,creatinine,urea nitrogen,24 hours urine protein and TIMP-1 expression in kidney of rats in sericin treatment group,sericin prevention group and metformin group were significantly lower than that of model group （P0.01,P0.05）.Moreover,there was no obvious differences between sericin treatment group,sericin prevention group and metformin group （P0.05）.Conclusions Sericin can obviously improve the renal function and decrease TIMP-1 expression in kidney of DN rats,and has satisfactory protective and preventive effects on interstitial fibrosis and disorganization of kidney glomeruli.Moreover,the therapeutical and preventive effects of sericin on DN
