摘  要:番茄不孕病毒是福建省切花菊病毒病的主要病原病毒,平均带毒率高达85.9%。为探讨其脱毒技术及其可行性,以福建省种植的菊花品种为试验材料,采用①热处理(白天40℃、夜间30℃,5~20d),②热处理结合茎尖培养,③热处理结合茎尖培养再二次培养等3种处理,经生物学鉴定、电镜观察和间接酶联免疫吸附试验检测(ID-ELISA),3种处理的脱毒率分别为13.3%~20.0%、41.6%~60.0%和83.3%~90.0%。此外,还进行了不同品种和外植体长度对切花菊组织培养的诱导率及脱毒率的影响试验。结果表明,热处理(白天40℃、夜间30℃,10~20d)结合茎尖培养(取茎尖0.5~1.0mm)再二次培养是脱除切花菊的番茄不孕病毒的最有效方法,可在大田生产中应用推广。Tomato aspermy virus (TAV) is a main chrysanthemum virus in Fujian,with an average infection rate of 85 9%.In order to study the virus eliminating technique and its feasibility,the experiment was conducted with the chrysanthemum planting in Fujian as material and designed as 3 treatment,i.e.,heat treatment (day 40℃ and night 30℃,5-20 d),shoot tip culturing with preheat treatment and secondary tissue culturing with preheat treatment.The results of biological identification,electron microscopy and ID ELISA test showed that the elimination efficiencies of TAV were 13 3%-20 0%,41 7%-60 0% and 83 3%-90 0%,respectively.The effect of various cultivars and explant length on induced rate and eliminating rate was studied.The results showed that secondary tissue(0 5-1 0 mm shoot tip) culturing with preheat treatment (day 40℃ and night 30℃,10-20 d) was the most effective way in elimination of TAV.It can be applied and popularized in the field production.
