摘 要:目的观察不同剂量的黄芪多糖(APS)对糖尿病大鼠胰腺Kir6.2表达的干预作用。方法建立Wistar大鼠糖尿病模型,随机分为模型组、对照组、APS低剂量干预组和APS高剂量干预组,进行糖耐量实验(OGTT),检测胰腺Kir6.2mRNA和蛋白表达变化。结果模型组OGTT30min、60min、120min和180min血糖值显著高于对照组(P<0.05),而APS高剂量干预组的60min和120min血糖值较模型组显著降低(P<0.05);与对照组比较,模型组胰腺Kir6.2mRNA和蛋白表达水平显著下降(P<0.05),APS低剂量干预组和APS高剂量干预组Kir6.2的mRNA和蛋白表达水平较模型组有不同程度上调(P<0.05)。结论黄芪多糖对糖尿病大鼠胰腺Kir6.2表达有一定的上调作用,可能是其发挥降糖作用的机制之一。Objective To investigate the effects of Astragalus polysaccharide(APS) on the expression of pancreatic Kir6.2 in diabetes rats.Methods Wistar rats were divided into model group,control group,low-dose APS group and high-dose APS group.Oral glucose tolerance test(OGTT) was performed,and Kir6.2 mRNA and protein expression in pancreas were performed by RT-PCR and Western blot.Results In OGTT,the blood glucose levels in model group at 30 min,60 min,120 min and 180 min were significantly higher than those in control group(P〈0.05),but in high-dose APS group the blood glucose levels at 60 min and 120 min were significantly lower than those in model group(P〈0.05);the expression of Kir6.2 in model group decreased significantly,and the expressions of Kir6.2 were higher in low-dose APS group and high-dose APS group than in model group(P〈0.05).Conclusion Astragalus polysaccharide can up-regulate the expression of pancreatic Kir6.2 to reduce blood glucose.
