摘  要:将西伯利亚鲟分别经腹腔注射接种嗜水气单胞菌和浸泡在菌浓度为1×107CFU/mL的水体中,腹腔注射组分别于接种后第12、24和36h剖杀,浸泡组分别于浸泡第24、48和72h剖杀,采用免疫组织化学方法检测嗜水气单胞菌在西伯利亚鲟体内的分布。结果显示,注射组在第12h阳性信号首先出现在脾、肾、肝、心肌中;第24h在肠道、肌肉、胃及性腺中出现阳性细胞。浸泡组第24h阳性信号首先出现在脾、肾、肝、心肌中;第48h在肠道、肌肉、胃及性腺中存在阳性细胞。阳性信号主要位于感染细胞的胞质中,偶尔出现在组织间质中。试验证实,嗜水气单胞菌首先侵袭的是脾、肾、肝、心肌、肠道等组织,然后是肌肉和性腺,而脑组织中一直未检测到阳性细胞。Acipenser baeri was inoculated intraperitoneally with Aeromonas hydrophila at 0.5mL/tail or immersed in the water with concentration of the bacteria at 1×107CFU/mL respectively.The subjects in intraperitoneally-inoculated group were slaughtered at hours 12,24,and 36 after inoculation,respectively,while the subjects in immersed-group were slaughtered at hours 24,48 and 72 after inoculation respectively.Tissues were collected for preparing tissue section and immunohistochemistry protocal were developed to detect the distribution of the pathogen in samples.The results showed that the positive signals appeared firstly in myocardium,spleen,kidney and liver in the 12h-inoculated group,and then presented in gut,muscle,stomach and gonad in the 24h-inoculated group.The myocardium,spleen,kidney,liver were also positive for the pathogens in the 24h-immersed group,and positive signals presented in the gut,muscle,stomach and gonad in the 48h-immersed group.Positive signals were mainly located in the cytoplasm of infected cells,and occasionally appeared in the connective material between the tissues.The results showed that A.hydrophila firstly invade the myocardium,spleen,kidney,liver,intestines and other organs,and then spreaded to the muscles and gonads;while positive signals were not detected in the brain tissue.
