气虚血瘀证与Balb/c小鼠结肠癌血行转移相关性的机理研究  被引量:5

摘  要:目的:通过建立气虚血瘀证Balb/c小鼠结肠癌人工血行转移的复合模型,研究气虚血瘀证对Balb/c小鼠结肠癌人工血行转移的影响及其相关机制。方法:将36只8周龄雌性Balb/c小鼠随机分为6组,即空白对照组、血瘀组、肿瘤A组、肿瘤B组、瘀加瘤A组和瘀加瘤B组。第28天处死空白对照组、血瘀组、肿瘤A组和瘀加瘤A组,检测血黏度、ERK1/2、VEGF。对肿瘤B组和瘀加瘤B组,进行生存期观察。结果:血瘀组及瘀加瘤组全血黏度均明显升高,与空白对照组比较差异极显著(P<0.01)。瘀加瘤组右肺转移灶计数明显高于肿瘤组,差异有显著性(P<0.05)。生存期短于肿瘤组,差异有统计学意义(P<0.05)。ERK1/2、VEGF按血瘀组、肿瘤组、瘀加瘤组顺序梯度递增,与空白对照组比较差异显著(P<0.05)。结论:气虚血瘀证能促进结肠癌转移,缩短其生存期。气虚血瘀证可能是通过上调ERK1/2、VEGF的表达,从而促进结肠癌细胞的增殖和转移。Objective:To Study the mechanism of relationship between the qi deficiency and blood stasis syndrome and artificial hematogenous metastasis of colon cancer by establishing a compound model of qi deficiency and blood stasis syndrome and artificial hematogenous metastasis of colon cancer.Methods:Thirty six eight-weeks old female Balb /c rats were divided into six groups randomly:control group,stasis group,tumor groupA B,stasis and tumor groupA B. We observed the survival time of tumor groupB,ERK1/2 and VEGF At the 28th day,after killing the mice of the other groups,we detected the whole blood viscosity and the amounts of metastasis tubercle on lung. Results:The whole blood viscosity of stasis group,stasis and tumor group was more significantly improved than control group(P〈0.01). The amounts of metastasis tubercle on lung in stasis and tumor group were more significantly improved than tumor group(P〈0.05). The survival time of stasis and tumor group was significantly shoter than tumor group(P〈0.05). The protein expression of ERK1/2,VEGF increased in order of stasis group,tumor group,stasis and tumor group which was significantly strengthened than control group (P〈0.05).Conclusion:The influence of qi deficiency and blood stasis syndrome can promote the hematogenous metastasis of colon cancer. The mechanism is probably related to the enhancement of the expression of ERK1/2 VEGF.
