摘  要:目的:通过通心络胶囊对载脂蛋白E基因敲除[(ApoE(-/-)]模型组小鼠血清血栓素B2(TXB2)、6-酮-前列腺素F1α(6-Keto-PGF1α)指标的干预变化,探究其防治冠心病(CHD)的机理。方法:将40只ApoE(-/-)小鼠作为实验组,建立高脂血症和冠状动脉粥样硬化(AS)模型,随机分为模型组等5组,分别给予辛伐他汀及不同剂量的通心络胶囊进行干预,另选8只同系的小鼠作为正常对照组。通过对模型组小鼠血清TXB2、6-Keto-PGF1α水平的观察,分析通心络胶囊对ApoE(-/-)小鼠冠状AS的调节作用和机理。结果:模型组ApoE(-/-)小鼠血清TXB2含量高于正常组,6-Keto-PGF1α含量低于正常组,与正常组相比均有统计学意义(P<0.05);通心络胶囊干预后,通心络胶囊高剂量组和辛伐他汀组与模型组相比,血清TXB2、6-Keto-PGF1α含量差异均有统计学意义(P<0.05);通心络胶囊高、中、低剂量组与辛伐他汀组相比,差异均无统计学意义(P>0.05)。结论:通心络胶囊能降低ApoE(-/-)小鼠血清TXB2、升高6-Keto-PGF1α水平,表明通心络胶囊具有抗凝,抑制血栓形成,达到恢复内皮功能,改善心肌缺血损伤,防治CHD的作用。Objective:by measuring the index changes of the serum thromboxane B2(TXB2)and 6-Keto-PGF1α of Apolipoprotein E gene knockout mice[ApoE(-/-)] intervened by Tongxinluo Capsule,then to explore the effect and mechanism of Tongxinluo Capsule regulating coronary heart disease(CHD).Methods:Forty Apolipoprotein E gene knockout mice[ApoE(-/-)] were taken as the experimental group,to establish the models of hyperlipidemia and coronary atherosclerosis,then randomly divided to 5 groups by respectively given Simvastatin and different doses of Tongxinluo Capsule to intervene.Other 8 isogeneic mice were chosen for the normal control group. Analysis on the effect and mechanism of Tongxinluo Capsule regulating coronary atherosclerosis by researching the level of TXB2 and 6-Keto-PGF1α of ApoE(-/-)mice of Model groups.Results:The amount of TXB2 of ApoE(-/-)mice in model groups is more than in normal group,while the amount of 6-Keto-PGF1αis less,which all have statistical significance comparing with normal group(P〈0.05).After being intervention by Tongxinluo Capsule,the amount of TXB2 and 6-Keto-PGF1α in high dose of Tongxinluo group and Simvastatin group are different with which in Model group(P〈0.05),while the difference among Simvastatin group and all the doses of Tongxinluo groups have no statistical significance(P〉0.05).Conclusion:Tongxinluo Capsule can decrease TXB2 and 6-Keto-PGF1α of Apolipoprotein E gene knockout mice[ApoE(-/-)],that's to say Tongxinluo Capsule can anticoagulation,inhibiting thrombosis,recover the endothelial function and improve myocardial ischemia,then prevent and regulate CHD.
