摘 要:目的观察愈灵颗粒对戊四氮(PTZ)致大鼠海马纤维出芽(MFS)的影响。方法选取70只清洁级SD大鼠,从中随机选取10只作为空白对照组。其余动物用戊四氮(PTZ)造模,将符合模型标准的大鼠随机分为3组,即愈灵颗粒组、丙戊酸钠组和模型组。运用Timm染色法观察MFS,探讨愈灵颗粒对海马MFS的影响。结果模型组与空白组比较,大鼠海马IML出芽目测评级差异无统计学意义(P>0.05),海马CA3区神经出芽的黑染点数目和面密度、平均光密度和积分光密度结果比较差异均有统计学意义(P<0.05),综合常规切片结果提示癫可导致海马神经元死亡,进而导致纤维出芽。愈灵颗粒组与模型组比较,愈灵颗粒可降低海马CA3区神经出芽的数目,黑染点数目和面密度、平均光密度和积分光密度结果比较差异均有统计学意义(P<0.01)。结论①致大鼠存在海马MFS现象;②愈灵颗粒可以减少癫大鼠海马MFS的数量和降低阳性程度,说明愈灵颗粒可保护海马神经元,抑制其纤维出芽。Objective To observe the effect of Yuxianling granule(YXLG) on the MFS in epileptic model rats caused by PTZ.Methods 70 SD rats,except for 10 rats were selected randomly as the blank control group,were modeled by pentylenetetrazol(PTZ) and divided randomly into three groups,YXLG group,sodium valproate group and model group.MFS was observed with Timm and the effect of YXLG on MFS was studied.Results Compared the model group,the difference of with blank group,the MFS of IML was not significant(P0.05) and the differences of the dyed-black points number and the dyed-black points'area density of MFS in CA3 of hippocampal slice were significant statistical significant(P0.01);Comprehensive routine biopsy results suggested that epilepsy could led to death of hippocampal neurons,and caused MFS.Compared YXLG group with model group,the differences of dyed-black points number and the dyed -black points'area density of MFS in CA3 were significant statistical significant(P0.01).Conclusions ① There exists the state of MFS in hippocampal slice.② YXLG can reduce the numbers and positive of hippocampal MFS,which suggests that YXLG can protect the hippocampal neurons.and inhibit MFS.
