摘  要:目的探讨柯萨奇-腺病毒受体(CAR)基因-968G/A多态性与中国南方汉族人群柯萨奇病毒性心肌炎的关系。方法收集病毒性心肌炎患儿280例,将其中CVB-IgM阳性130例作为心肌炎组,并选择同期130名健康儿童作为对照组,采用聚和酶链反应-限制性内切酶片断长度多态性(PCR-RFLP)、DNA测序等方法检测2组CAR基因-968G/A多态性,采用酶联免疫吸附法(ELISA)测定两组血清CAR水平。结果心肌炎组GG基因型和G等位基因频率均显著高于对照组(P<0.05)。心肌炎组血清CAR水平明显高于对照组(P<0.01),CAR基因-968G/A多态位点中含G等位基因(GG+GA)者血清CAR水平和CVB感染率均显著高于非G等位基因(AA)携带者(P均<0.05)。结论 CAR基因-968G/A多态性可能与中国南方汉族人群柯萨奇病毒性心肌炎有关,G等位基因是柯萨奇病毒性心肌炎的易感基因。Objectives To investigate the relationship between polymorphism of Coxsackie-adenovirus receptor(CAR) gene G968A and Coxsackie viral myocarditis in Han Population of South China.Methods 130 Coxsackie virus B-IgM positive patients(myocarditis group) derived from 280 patients with viral myocarditis and 130 healthy children(control group) were enrolled.The 968G /A polymorphism in CAR gene was detected by polymerase chain reactionrestriction fragment length polymorphism and DNA sequencing in two groups.Serum CAR levels in the two groups were determined by enzyme-linked immunosorbent assay.Results The frequencies of CAR GG genotypes and G alleles in myocarditis group were significantly higher than those in control group(P〈0.05).Serum CAR level in myocarditis group was significantly higher than that in control group(P〈0.01).The people with CAR 968G allele(GG + GA) had higher level of serum CAR and Coxackie virus B infection rate than those only with CAR 968A allele(AA)(P〈0.05).Conclusions CAR gene-968G /A polymorphism is associated with Coxsackie viral myocarditis in the Han population of South China,and G allele is a susceptible gene of Coxsackie viral myocarditis.
