摘  要:目的了解静态负荷对骨骼肌细胞线粒体的影响。方法 96只健康雄性Wistar大鼠,随机分为12组,每组8只。将大鼠麻醉后游离其下肢比目鱼肌远端,分别施加0、25、50和100g负荷,各负荷组持续时间分别为30、60和90min,在4℃条件下,9000r/min离心20min分离线粒体,测定比目鱼肌线粒体钙浓度、Ca2+-Mg2+-ATPase活力及丙二醛(MDA)水平。结果与对照组比较,大鼠骨骼肌线粒体钙浓度在负荷25g组负荷90min,线粒体钙浓度增加(P<0.05);50和100g组负荷60、90min,线粒体钙浓度增加(P<0.05)。线粒体Ca2+-Mg2+-ATPase活力在负荷100g组90min降低(P<0.05);线粒体MDA水平随负荷增大和负荷时间延长而增加。结论长时间静态负荷作用于骨骼肌,可引起细胞线粒体钙浓度增高,Ca2+-Mg2+-ATPase活力降低,MDA水平增加。Objective To study the influence of static load on mitochondria of skeletal muscle.Methods 96 male Wistar rats were randomly divided into 12 groups of 8.After the rats were anesthetized,the distal soleus at the far end of the lower limbs were dissociated and respectively loaded with 0,25,50 and 100 g for 30,60 and 90 min.Mitochondria was separated by centrifuge at 9 000 r/min for 20 minutes at 4 ℃.Then observed were the concentration of Ca2+ in mitochondria of soleus,the activity of Ca2+-Mg2+-ATPase and the level of MDA in mitochondria.Results Compared with the control groups,the concentration of Ca2+ in mitochondria of skeletal muscle significantly increased in the groups with loads of 25 g for 90 min (P0.05).It also increased in the groups with loads of 50 g for 60 min and 90 min and 100 g for 60 min and 90 min,showing statistical significances (P0.05).The activity of Ca2+-Mg2+-ATPase in mitochondria decreased in the groups with loads of 100 g for 90 min,which was statistically significant (P0.05).The level of MDA in mitochondria increased with the loads added and time of exertion prolonged.Conclusion Long time of static loads affects physiological function of mitochondria through the increase of Ca2+ concentration,the decrease of Ca2+-Mg2+-ATPase activity and the increase of MDA level.
