摘 要:目的观察冰黄液对大鼠急性脑出血后脑水肿的防治作用并探讨其作用的可能机制。方法应用胶原酶和肝素混合液脑内注射诱导大鼠急性脑出血模型,分别采用干湿质量法、生物发光法以及放免法测定术后6h及第1、3、5天各组大鼠的脑组织含水量以及血肿周围脑组织三磷酸腺苷(ATP)、内皮素-1(ET-1)的水平。结果与假手术组大鼠相比,脑出血模型组的大鼠脑组织含水量自术后6h即显著增高,术后72h达高峰,脑组织内ATP含量自术后24h开始呈持续性降低,与此相对应,脑出血后6h血肿周围脑组织内ET-1的含量也显著升高,至术后第3天达最高值,其变化与脑水肿程度存在明显的正相关。给予大鼠冰黄液治疗可以显著减轻大鼠脑水肿程度,同时降低局部脑组织内ET-1含量,提高脑组织内ATP水平。结论冰黄液治疗对大鼠脑出血后脑水肿具有较好的防治作用,其作用与减少脑出血后局部脑组织内ET-1生成,缓解脑出血后血管痉挛,改善脑出血后局部脑组织的能量代谢障碍等机制密切相关。Objective:To study the effect and the mechanism of Binghuang Decoction on brain edema of intracerebral hemorrhage in rats.Methods:Rats cerebral hemorrhage was induced by injecting 2μl mixed liquids.The contents of water,ATP,and ET-1 of brain tissue around hematoma at 6hr,24hr,72hr and 120hr were detected by the dry-wet-weighting technique,the bioluminescence method and the radioimmunoassay method.Results:Compared with the shame operation group,the contents of water in cerebral hemorrhagic rats were raised obviously since 6hr after operation and reached maximum at 72hr,while the ATP contents were decreased since 24hr continuously.Correspondingly,the ET-1 contents were also increased and correlated positively with the increases of the water contents.In contrast,the contents of water and ET-1 were significantly lower and the content of ATP was significantly higher in the Binghuang Decoction group.Conclusion:Binghuang Decoction has a protection function on cerebral hemorrhagic edema which is related to the production of ET-1,remove cerebrovascular spasm and improve the energy metabolism.
