摘 要:目的 研究流产孕妇不同乙型肝炎病毒脱氧核糖核酸(HBV-DNA)含量对外周血淋巴细胞染色体着丝粒点(Cd)变异的影响,探讨孕妇自然流产原因.方法 选择有自然流产史孕妇60例,根据HBV-DNA含量,分为2组[其中Ⅰ组HBV-DNA含量为103~105 copy/mL;Ⅱ组HBV-DNA含量大于105 copy/mL];选取15例健康体检者作为健康对照组.对上述各组对象进行外周血淋巴细胞染色体制作,同步银染法观察染色体着丝粒点变异情况,分析变异率与HBV-DNA含量的关系.结果Ⅰ组与健康对照组、Ⅰ组与Ⅱ组之间着丝粒点变异率差异均有统计学意义,Ⅱ组与健康对照组着丝粒点变异率有极显著性差异,且病毒含量越高,变异率越大.结论 乙型肝炎病毒可引起外周血染色体着丝粒点变异,使着丝粒点功能失活,影响纺锤丝与着丝粒的附着,从而导致染色体不分离,提示乙型肝炎患者Cd的变化可能是导致流产的重要原因之一.Objective To study the impact of HBV-DNA on the variation of peripheral blood lymphocytes chromosome centromere, to explore the causes of spontaneous abortion in pregnant women. Methods 60 women with history of spontaneous abortion were selected, according to HBVDNA content, they were divided into 10 3 -10 5 (I group)and〉10 5 (II group)groups; 15 cases selected as healthy control group. The peripheral blood lymphocytes chromosome was prepared and simultaneous observation of silver staining chromosome centromere-point variation of mutation rate and analysis the relationship between HBV-DNA content. Results There were significantly different between I group and the control group,I group and II groups There was significant difference,between II group and the control group, and the higher the viral load, the greater the mutation rate. Conclusion Peripheral blood of hepatitis B virus can cause chromosome centromere point mutation,so that the functional centromere inactivation influence the spindle wire attached to the centromere, resulted in chromosome nondisjunction. It suggest that changes in Cd hepatitis B patients may lead to abortion one of the important reasons.
