摘  要:应用植块法,比较大泷六线鱼(Hexagrammos otakii)脾脏细胞在2种基础培养基和3个胎牛血清浓度下的体外培养效果。根据细胞的迁出、贴壁、细胞种类以及生长状况,得出大泷六线鱼脾脏细胞适宜的体外培养体系为:MEM+15%胎牛血清、pH7.2-7.4、20℃;培养产物由6种细胞组成,其中C型细胞培养120 d以上依然状态良好,并进行了传代培养。培养启动12 d后,C型细胞层上出现造血灶,造血灶细胞最长可以维持50 d以上。本研究为大泷六线鱼脾脏细胞系的构建和脾脏功能的研究奠定了基础。In vitro culturing effect of Hexagrammos otakii splenic cells was compared by explant method,with two media(L-15 and MEM) and three serum concentrations(10%,15% and 20% fetal bovine serum,FBS).Based on the migrating,change the font to be the same as the other main part of the paragraph,the optimal culture system for H.otakii splenic cells was MEM medium supplemented with 15%FBS,at pH 7.2-7.4 and 20 ℃.Under this condition,six types of cells could migrate out of the splenic explant.Type C cells grew gradually into a confluent monolayer or multilayer and could be successful subcultured 120 days later.The hematopoietic foci which were formed by round cells closely associated to type C cells were observed 12 days later in MEM medium with three FBS concentrations.The foci could persist up to 50 days,and then declined in size and disappear.This work provided a useful foundation for the establishment of in vitro cell-line and the functional investigation in H.otakii spleen.
