反相波相色谱法快速测定尿羟脯氨酸

摘　　要：报道了以异硫氰酸苯酯为柱前衍生剂，采用反相液相色谱快速测定尿羟脯氨酸（HYP）的方法。该法的线性范围为31．25～500μmol／L，相关系数0．996，批内、批间的变异系数为2．3％和3．3％，回收率为95．1％，同时测定了68例60～85y老年人尿羟脯氨酸的含量（参考值以尿HYP含量与尿肌酚比值计算），老年女性的结果（315．4±97．0μmol／g．Cr）显著高于老年男性（198．2±78．7μmol／g．Cr），P＜0．01。A rapid method was described for the determination of urianry hydroxyproline (HYP) by reversed phase high performance liquid chromatography. After acid hydrolysis of urine, HYP was derived with phenylisothiocyanate (PITC). Calibration curves for HYP was linear from 31. 25 to 500 μmol/L , the correlation coefficients between the peak area of HYP and HYP concentration were 0. 996, within-assay CV was 2, 3% and the between-assay CV was 3.3 %. Analytic recovery of HYP added to urine samples was 95. 1 %. Measurement was made on 68 old people aged 60~80yearsold. The mean value of old women (315. 4±97. 0 μmol/g. Cr) was higher than that of old men (198. 2±78. 7 μmol/g. Cr). P<0. 01.
