摘  要:目的:探讨川芎嗪对TNF-α诱导的HBMEC细胞MCP-1mRNA的影响及p38MAPK的作用。方法:HB-MEC细胞培养,加入TNF-α,采用逆转录-聚合酶链反应测定(reverse transcription-polymerase chain reaction,RT-PCR)检测MCP-1 mRNA的水平;免疫印迹法检测磷酸化p38的表达。川芎嗪预处理1h,入TNF-α检测变化。结果:TNF-α浓度依赖地增高MCP-1表达;川芎嗪组MCP-1减少;TNF-α组p-p38增加,川芎嗪组较TNF-α组减少。结论:TNF-α可能通过p-p38介导HBMEC细胞MCP-1 mRNA的表达;川芎嗪可能通过抑制p-38通路抑制MCP-1的表达。Objective:To investigate the effect of tetramethyl pyrazine on MCP-1 mRNA expression in HBMEC cells induced by TNF-αand the effect of p-38 MAPK.Methods:HBMEC cells were cultured.Concentration of TNF-αused to treat the HBMEC cells was selected.The mRNA expression of MCP-1 HBMEC cells was detected by reverse transcription-polymerase chain reaction(RT-PCR),p-38 MAPK expression was investigated by Western blot.Results:The mRNA expression of MCP-1 was increased when cells were exposed to different concentration of TNF-α;the p-p38 was increased,Tetramethyl pyrazine could inhibit the increase of MCP-1 mRNA.The expression of p-p38 was decreased.Conclusion:MCP-1 mRNA was increased after TNF-αexposure;Tetramethyl pyrazine could inhibit MCP-1 mRNA expression via p38 MAPK.
