摘　　要：目的 研究氯沙坦对血管内皮细胞凋亡的影响.方法 体外培养原代人脐静脉内皮细胞（human umbilical vein endothelial cell,HUVEC）,培养成功后先用血管紧张素Ⅱ（Ang Ⅱ）刺激细胞,然后再加入氯沙坦与Ang Ⅱ共同作用于HUVEC,采用流式细胞术A-V双染法观察细胞的凋亡率.结果 HUVEC加入Ang Ⅱ刺激24 h后,细胞凋亡率较正常对照组明显增加,10^-7 mol/L Ang Ⅱ组、10^-6 mol/L Ang Ⅱ组、10^-5 mol/L Ang Ⅱ组的细胞凋亡率分别是7.85%、9.96%和10.4%;加入10^-6 mol/L氯沙坦后,HUVEC凋亡率得到明显改善,细胞凋亡率分别为7.95%、8.62%、8.67%.结论 氯沙坦对Ang Ⅱ诱导的HUVEC细胞凋亡有明显的抑制作用.Objective To study the effects of losartan on vascular endothelial cell apoptosis. Methods Primary human umbilical vein endothelial cells （HUVEC） were cultured in vitro. The cultured cells were stimulated with angiotensin Ⅱ （Ang Ⅱ ） , and then losartan was added to act on HUVEC together with Ang Ⅱ . HUVEC apoptosis rate was measured by flow cytometry. Results After Ang Ⅱ stimulation for 24 hours, HUVEC apoptosis rate significantly increased compared with normal control group. The apoptosis rate of three Ang Ⅱ groups （10^-7 mol/L, 10^-6 mol/L, 10^-5 mol/L） were 7.85%, 9.96% and 10. 4%,respectively. After adding 106 mol/L losartan,HUVEC apoptosis rates were 7.95%, 8. 62% ,8. 67% accordingly. Conclusions Losartan can inhibit HUVEC apoptosis induced by Ang Ⅱ.
