摘  要:采用改良氯化苄法、CTAB法、蛋白酶K裂解法这三种方法,对冬虫夏草菌丝体基因组DNA进行提取,将提取的基因组DNA经过紫外分光光度计检测纯度和计算浓度、琼脂糖凝胶电泳分析及PCR扩增,结果表明,3种方法均能提取到符合分子生物学的DNA,其中蛋白酶K裂解法纯度最高,含蛋白质少,改良氯化苄法、CTAB法次之。而浓度则是改良氯化苄法最高,CTAB法、蛋白酶K裂解法次之。3种方法所提取的DNA均能扩增出理想条带。Three methods of modified benzyl chloride method,CTAB method and proteinase K lysis method were used to extract the DNA of cordyceps mycelium,the concentration of DNA was detected by UV spectrophotometer and calculated the DNA purity,agarose condensate gel electrophoresis and PCR amplification was analyzed.The results showed that,the three methods were able to extract the DNA from Cordyceps mycelium,the DNA which was extracted by proteinase K lysis method was the highest purity,containing less protein and the modified benzyl chloride method and the CTAB method followed.The DNA which was extracted by the benzyl chloride method was the highest concentration,the CTAB method followed,then the proteinase K lysis method.The DNA which was extracted by three kinds of method could amplify the ideal bands.
