摘 要:为了解明培养基中添加的植物生长调节剂噻二唑苯基脲(TDZ)对桑树愈伤组织诱导和促进不定芽分化的作用机制,以陕桑305组培苗为试验材料,检测在离体叶片培养的过程中TDZ对细胞膜透性、过氧化物酶(POD)活性等生理指标的影响。当桑树叶片离体培养45d时,经TDZ处理后的叶片电导率、丙二醛(MDA)含量、POD活性和叶绿素含量分别为对照的74.8%、73.9%、68.3%和180.8%。研究结果表明TDZ可通过抑制离体培养桑树叶片的脂质过氧化作用,保护细胞膜结构的完整性;可通过抑制叶片中POD的活性,阻止叶绿素的降解,延缓叶片的衰老,从而提高离体培养桑树叶片的再生效率。In order to reveal the functioning mechanism of plant growth regulator Thidiazuron (TDZ) in culture medium on mulberry callus induction and adventitious bud differentiation,the cultured seedlings of mulberry Shansang 305 was used to study the effects of TDZ on cell membrane permeability,peroxidase (POD) activity and other physiological properties to leaves cultured in vitro.The results showed that,at the 45th day after in vitro culture of mulberry leaves,the e-lectrical conductivity,malonaldehyde (MDA) content,POD activity,and chlorophyll content in TDZ-treated leaves was 74.8%,73.9%,68.3%,and 180.8% as much as that in the control leaves,respectively.These results suggest that TDZ could protect the integrity of cell membrane structure through inhibiting peroxidization of lipids in mulberry leaves cultured in virto.Meanwhile,it could inhibit POD activity in the leaves,prevent degradation of chlorophyll,and delay senility of in vitro leaves,thus promoting the regeneration efficiency of mulberry leaves cultured in vitro.
