RAPD Analysis of Genetic Relationship among Three Populations of Chilean Jack Mackerel(Trachurus murphyi) from Southeast Pacific

摘  要:利用RAPD分子标记技术对智利外海3个群体(IEZ1、OEZ1和OEZ2)的53条竹筴鱼进行群体遗传结构及遗传多样性的分析。研究结果表明:在可供试验引物中筛选到具有多态性的RAPD引物3个。每个引物扩增1-7个可辨认片段,3个随机引物在竹筴鱼3个群体中共检测到13条带片断,其中9个(69.2%)片断显多态性。OEZ1的多态性最高,达23.08%,IEZ1、OEZ2的多态性为15.38%。经济区内外的群体多样性差别不大。IEZ1的Shannon信息指数为0.0930,OEZ1为0.1396,OEZ2为0.0930。IEZ1 Nei’s基因多样性为0.0637,OEZ1为0.0956,OEZ2为0.0637。Nei’s遗传距离表明IEZ1与OEZ1最大,IEZ1与OEZ2稍小,OEZ1和OEZ2最小。Shannon信息指数和Nei’s遗传距离表明,经济区外的2批群体与经济区内的群体遗传关系差距并不显著。The genetic structure and genetic diversity of the three populations of Chilean jack mackerel Trachurus murphyi collected from inter-EEZ and outer-EEZ coast off Chile in 2005 and 2006 were analysed by RAPD-PCR markers.Three polymorphic primers were chosen among the experimental primers.There were 13 bands in the three populations by RAPD-PCR with three 3-base primers,and 1~7 bands were detected for one primer.Nine primers were polymorphic,accounting for 69.2%.The group OEZ1 had the maximal polymorphism of 85.19%,followed by groups IEZ1 OEZ2,with the polymorphism of 15.38%.There were no significant differences in genetic diversity among the three groups.POPGENE software revealed that Shannon's Information index of IEZ1 and OEZ2 were 0.0930,while that for OEZ1 was 0.1396.Nei's gene diversity of IEZ1 and OEZ2 were 0.0637,while that for OEZ1 was 0.0956.There was the maximum genetic distance between IEZ1 and OEZ1,followed by IEZ1 and OEZ2,and the minimum for OEZ1 and OEZ2.The Shannon's Information index and Genetic Distance indicated that the difference in genetic diversity between inter-EEZ group and outer-EEZ group was not significant.
