摘 要:用小鼠肝脏离体灌流和大鼠肝脏组织切片培养方法,观察了甲醛对肝脏多种酶、谷胱甘肽(GSH)及细胞亚微结构的影响。结果表明:甲醛致乳酸脱氢酶(LDH)活性增加,5'-核苷酸(5'-N)和葡萄糖-6-磷酸酶(G-6-P)活性下降,GSH含量下降,细胞膜和内质网亚微结构的损伤,且均存在良好的剂量相关效应。本文结果提示,甲醛的肝脏毒性不仅对其本身毒作用,而且对物质代谢和其他毒物的代谢与作用有重要意义。The activity of lactate dehydrogenase (LDH) in the medium of perfused mice livers was increased 0.25- 14.0 mmol/L by formaldehyde. Enzyme histochemistry showed inhibition of 5'-nucleotidase (6'-N) and glucose-6-phosphate dehydrogenase (G-6-P) activity. 5'N is a marker for cell membranes function and its structural integrety. Decreased activity of this enzyme suggests abnormality of membrane and whole cell function. G-6-P may be taken as an indicator of cell metabolism. Gaps in the cell membrane and on smooth surfaced endoplasmio reticula were seen under eleotronmioroscopie observation. These also indicate adverse effects of formaldehyde on the liver.In the cultured liver lides, glutathione (GSH) was significantly decreased. GSH may be involved in many biological functions, especially in the biotransfor-mat.ion of chemical toxicants. Therefore, exposure to formaldehyde may increase the toxicity of these GSH-requiring chemicals.
