摘 要:以卡特兰Cattleya labiata品种MC1类原球茎(Protocorm-like bodies,以下简称PLBs)为外植体,分别接种于改良VW+2.0 mg/L6—BA+0.2 mg/L NAA+100.0 g/L香蕉泥+不同浓度的乙烯抑制剂(AgNO3或CoCl2)的增殖培养基以及改良VW+2.0 mg/L6—BA+0.05 mg/L NAA+100.0 g/L香蕉泥+不同浓度AgNO3的长叶培养基中研究PLBs的增殖与分化情况。结果表明:促进卡特兰PLBs增殖的最佳AgNO3浓度是10.0~20.0 mg/L,最佳CoCl2浓度是10.0 mg/L;AgNO3还有促进PLBs分化的作用,其最佳浓度是1.0 mg/L。PLBs经分化长叶培养基与生根壮苗培养基分化培养后,获得了大量生长健壮的组培小苗,并移栽成活。以卡特兰的PLBs和组培小苗幼叶为材料进行石蜡切片,在光学显微镜下观察其内部形态结构。结果表明核质比大、分裂能力大的细胞集中分布在PLBs的外周部分。PLBs从幼到老的生长过程中,其外表面颜色由浅到深,外周分裂能力强的细胞其数目大量增加。Protocorm-like bodies(PLBs) of Cattleya MC1 were inoculated on the propagation medium of the modified VW containing 2.0 mg/L 6-BA,0.2 mg/L NAA,100.0 mg/L banana mud,ethylene inhibitor(AgNO 3 or CoCl 2) with different concentration,and on regeneration medium of the modified VW containing 2.0 mg/L 6-BA,0.05 mg/L NAA,100.0 mg/L banana mud,and AgNO 3 with different concentration.The results showed the best concentration of AgNO 3 for promoting propagation of Cattleya PLBs is 10.0~20.0 mg/L,the best concentration of CoCl 2 is 10.0 mg/L.AgNO 3 also could promote regeneration of PLBs,the best concentration was 1.0 mg/L.PLBs were differentiated into many plantlets on regeneration buds medium and rooting medium and transplanted successfully into matrix.The protocorms(PLBs) and young leaf of Cattleya were studied by paraffin section method and observed under optical microscope.The result showed the cells with high ratio of nucleus to cytoplasm and high division ability were at the outer layer of PLBs.In the process of PLBs growing up from young to old,facial color of PLBs were becoming deeper,and the number of the cells with high division ability at the edge of PLBs increased largely.
