摘  要:目的研究SUMO-1在低氧激活的HIF-1/VEGF信号通路中作用。方法构建稳定表达SUMO-1的细胞系,采用荧光显微镜观察SUMO-1的细胞内定位,利用Western blot检测在低氧培养细胞中SUMO-1和HIF-1α的表达;利用荧光素酶报告基因实验检测SUMO-1对依赖于HIF-1的报告基因表达的影响。结果 SUMO-1主要分布在细胞核中,低氧可以上调SUMO-1的表达,SUMO-1可以上调HIF-1α的表达,促进依赖于HIF-1的荧光素酶报告基因的表达。结论低氧可能通过上调细胞中SUMO-1的表达,进而稳定HIF-1α或者上调HIF-1α的表达,增强HIF-1的转录活性。Objective To evaluate the role of SUMO-1 in hypoxia activated HIF-1/VEGF signaling pathway. Methods The recombinant plasmid expressing GFP-SUMO-1 was transfected into HER293 cells, and the positive clones were screened by G418 to establish the stable expression cell line. Fluorescence microscope was used to observe the distribution of overexpression of SUMO- 1 protein in HER293 cells. Western blotting was used to elucidate the expression of SUMO-1 and HIF-1α during hypoxia, and the luciferase reporter gene assay was used to evaluate the effect of SUMO-1 on the expression of HIF-1-dependent reporter genes. Results The distribution of SUMO-1 protein is mainly in nucleus of the transfected HER293 cells. Hypoxia upregulated the expression of SUMO-1 protein, and overexpression of SUMO-1 promoted HIF-1α expression and increased the expression of HIF-1-dependent reporter genes. Conclusion Upregulation of SUMO-1 proteins after hypoxic stimulation may stabilize HIF-1α or increase its expression and enhance its transcriptional activity.
