摘  要:[目的]研究黄芩苷对高糖诱导的人脐静脉血管内皮细胞株(CRL1730)细胞凋亡及细胞增殖抑制的影响。[方法]用不同浓度黄芩苷(0.1、0.2、0.4g/L)、D-葡萄糖(5.5、15、25、35、45、55mmol/L)共同作用培养的人脐静脉血管内皮细胞株(CRL1730)48h,流式细胞仪(flow eytometry,FCM)测定内皮细胞的凋亡率、细胞周期,噻唑蓝法(MTT法)测增殖率,酶联免疫吸附法(Enzyme-linked immunosorbent assay,ELISA)检测可溶性细胞间黏附分子-1(Serum soluble intercellular adhesion molecule-1,sICAM-1)、肿瘤坏死因子-α(Tumor necrosis factor-α,TNF-α),化学法检测一氧化氮(Nitric oxide,NO)水平,黄嘌呤氧化酶法检测超氧化物歧化酶(SOD)含量,并与正常糖浓度(5.5mmol/L)培养基组比较。[结果]正常糖浓度时,除MTT外,黄芩苷各浓度下指标无统计学差异。在使用高糖培养基时黄芩苷作用组凋亡率下降,G2M期细胞比例、MTT值明显高于单纯高糖组(P﹤0.05),并同时降低sICAM-1、TNF-α,使NO含量增加、SOD活力增高。[结论]黄芩苷对高糖导致的CRL1730细胞凋亡率的增加、细胞周期改变、增殖能力下降具有保护作用。其原因可能与其抑制炎症反应、氧化应激等因素有关。[Objective] To investigate the effect of Baicalin on proliferation and apoptosis in cultured human umbilical vein endothelial cells(CRL1730) exposed to high-level glucose. [Methods] The cultured CRL1730 exposed to different concentrations of glucose(5.5,15,25,35,45,55mmol /L) were treated with Baicalin(0.1、 0.2、 0.4g /L) for 48 hours. Proliferation and apoptosis of CRL1730 were evaluated by MTT assay and flow eytometry(FCM) ,respectively. The soluble intercellular adhesion molecule-1(sICAM-1) and tumor necrosis factor-α(TNF-α) were measured by ELESA. Nitric oxide was measured by colorimetry. The xanthine oxidase measured the activity of superoxide dismutase(SOD) ,which was compared with the culture medium with normal sugar content. [Results] Under the normal glucose concentration,there was no significant difference between different dose of Baicalin. Under the high glucose apoptosis rate of Baicalin groups declined,the proportion of G2M phase and proliferation of CRL1730 cells was significantly higher than that of no Baicalin group. At the same time,the sICAM-1 and TNF-α decreased and vitality of SOD and NO increased. [Conclusion] Under the condition of high glucose,Baicalin can correct the increase of apoptosis rate,abnormal change of cell cycle and the inhibition of proliferation. The cause is that Baicalin may inhibit inflammatory response and oxidative stress.
