明胶片菌种保藏法的应用

摘　　要：[目的]建立一种保存菌种时间长、使用方便,便于基层推扩应用的菌种保藏方法。[方法]于2004年9月,采用明胶片菌种保藏法保存了标准菌种ATCC26003金黄色葡萄球菌和ATCC27853绿脓杆菌。[结果]2005～2009连续5年,通过每年的传代鉴定,菌种的存活率100%且各项生化特性保存完好未发生变异或丢失。达到了预期良好目的,现继续保存观察中。[结论]该方法保存菌种时间长、使用方便又不需要昂贵的仪器设备,是一种特别适用于基层推扩应用的菌种保藏方法。[Objective] To build a convenient method of preserving bacterial species for a long time,which also can be easily popularized in a basic level unit. [Methods] During September 2004,Gelatin disk drying method was used to preserve standard strains of Staphylococcus aureus numbered ATCC26003 and Pseudomonas aeruginosa numbered ATCC27853. [Results] For consecutive five years from 2005 to 2009,subculture and identification were performed every year,the bacterial species survival rate reached to 100%,and these species retained its biochemical characteristics without variation or loss,which had achieved expectant purpose,and these bacterial species were still in observation. [Conclusion] Gelatin disk drying method is a convenient method without expensive instrument,which can preserve bacterial species for a long time and be easily promoted in a basic level unit.
