Inhibiting Lewis Tumor and Distribution of Lung Targeting Etoposide Chitosan-alginate Microsphere in Vivo in Mice

摘  要:目的研究制备的依托泊苷壳聚糖-海藻酸钠微球(Etoposide Chitosan-alginate Microsphere,ECAM)的体内抑瘤作用及体内分布。方法通过比较各组小鼠移植性Lewis肺癌的抑瘤率,考察含药微球的抑瘤效果;用高效液相色谱法测定各组织中依托泊苷浓度。结果依托泊苷微球抑瘤率为63.62%,明显高于其他各组;小鼠尾静脉注射微球后15min,肺内依托泊苷量占注入总量的(52.64±1.79)%。结论 ECAM具有明显的抑制移植性Lewis肺癌的作用和肺靶向效果。Objective To study the inhibiting Lewis tumor growth and distribution of etoposide Chitosan-alginate Microsphere in vivo in mice. Methods The anti-tumor effect of drug-containing microspheres was studied by comparing each group of mice transplanted Lewis lung cancer inhibition rate;The drug concent rations in various tissues were determined by high performance liquid chromatograph (HPLC). Results Etoposide microspheres inhibition rate was 63-52%,significantly higher than other groups.The ECAM was injected into mice and (52.64 ± 1.79)% of the total dosage was detected in lung tissue after 15rain from administration. Conclusion The ECAM showed obvious inhibition of transplanted Lewis lung cancer and remarkable targeting action to the lung.
