针刺对哮喘大鼠肺泡灌洗液SP-A表达的影响

摘　　要：目的：探讨针刺治疗哮喘的效应机制。方法：40只SD大鼠按计算机随机数字表分为空白对照组、生理盐水对照组（NS对照组）、哮喘模型组、哮喘模型针刺组（哮喘针刺组）、哮喘模型捆绑组（哮喘捆绑组）5组。哮喘针刺组取穴“大椎”“肺俞”“风门”进行针刺治疗；哮喘捆绑组只捆绑，不针刺；其他组不予干预。免疫印迹法（Western-Blot）检测肺表面蛋白A（SP-A）。结果：哮喘模型组的气道阻力在哮喘激发后3～6min时较空白对照组和NS对照组显著增高（均P〈0．01）。Western—Blot检测显示哮喘模型组大鼠支气管肺泡灌洗液（branchoalveolar lavage fluids，BALF）中SP—A表达水平比空白对照组、NS对照组大鼠显著降低（均P〈0．05），哮喘针刺组大鼠BALF中SP—A表达与哮喘模型组相比较显著升高（P〈0．05）。结论：针刺防治过敏性哮喘的机制与调节哮喘大鼠呼吸道内SP—A的表达有关。Objective To explore the mechanism of acupuncture for treatment of asthma. Methods Forty SD rats were randomly divided into 5 groups： blank control group, normal saline control group （NS control group）, asthma model group, asthma model with acupuncture group （asthma acupuncture group） and asthma model with binding group （asthma binding group）. The asthma acupuncture group was treated with acupuncture at ＂Dazhui＂ （GV 14）, ＂Feishu＂ （BL 13） and ＂Fengmen＂ （BL 12） ; the asthma binding group was only binding without acupuncture and no intervention was given in the other groups. Surfactant protein-A （SP-A） expression was examined by Western-Blot. Results Airway resistance in asthma model group was significantly higher than that in blank control group and NS control group from 3 to 6 min after asthma provocation （all P〈0.01 ）. Western-Blot detection showed that SP-A expression in bronchoalveolar lavage fluid （BALF） of the rats in asthma model group was significantly lower than that in blank control group and NS control group （both P〈0.05）, and which in asthma acupuncture group was significantly higher than that in asthma model group （P〈0.05）. Conclusion The mechanism of prevention and treatment of acupuncture for allergic asthma is related to regulating SP-A expression of airway in asthmatic rats.
