摘  要:目的 研究真皮多能干细胞移植对大鼠皮肤光老化(skin photoage)的治疗作用,为皮肤光老化提供新的治疗方法.方法 57只SD大鼠随机分为正常组8只和模型组49只,采用UVA和UVB紫外灯同时对模型组进行照射(2 h/d,连续照射60 d),随机取8只大鼠进行皮肤光老化模型鉴定,模型成功后随机分为对照组(不做任何处理)和治疗组(1×106细胞悬液).体外分离培养SD乳鼠DMSCs,经DAPI标记后,取治疗组大鼠9只,于皮肤光老化区真皮层内多点移植细胞悬液,分别在移植后1、3、7 d处死大鼠,在荧光显微镜下观察DAPI标记的DMSCs在皮肤光老化区域组织存活、迁移.其余16只大鼠在以同样的方法行DSMCs移植后于第4、8周分别采用免疫组织化学检测各组真皮中成纤维细胞的变化.结果 DMSCs移植后1、3、7 d,荧光显微镜观察到DAPI标记的阳性细胞在皮肤光老化区存活并逐渐向真皮层迁移;移植后4、8周,DSMCs治疗组大鼠光老化皮肤较对照组均有显著恢复;与对照组相比,移植4周后治疗组大鼠皮肤组织成纤维细胞增多,8周后广泛均匀弥散在皮肤各层,两组比较差异有统计学意义(P〈0.01).结论 DMSCs移植对大鼠皮肤光老化具有明显的治疗作用.Objective To investigate the therapeutical effect of dermal muhipotential stem cells transplantation on the skin photoage in rats in order to provide a novel treatment for skin photoage. Methods Fifty-seven SD rats were randomly divided into normal group (8 rats) and model group (49 rats). The skin photoage model of 49 rats was irradiated by using UVA and UVB uhraviolet light (2 h per day, 60 days continuous exposured), and 8 rats were randomly executed for the model identification of skin photoage. Then the rats of model group were randoraly divided into the control group for 16 rats and the treatment group for 25 rats ( 1 × 10 6 cell suspension). DMSCs from suckling SD rats were isolated and cultured in vitro, then labeled with DAP1. The labeled DMSCs were injected with multipoints into the photoage skin's dermal in the treatment group (9 rats) and they were sacrificed respectively at 1, 3 and 7 days after transplantation. The survival and nfigration of the transplanted DMSCs in the skin photoage dermal which showed positive DAP1 were observed by using fluorescence microscope. The other 16 rats in the treatment group were received DMSCs transplantation in the same way, immunohistochemistry was used to detect the changes of fibroblasts in dermal organizatian at 4 and 8 weeks after transplantation. Results The observation showed the DM- SCs labeled with DAPI were survival and migrated to derma. of skin photoage at 1,3 and 7 days after transplantation. Compared with the control group, the aging skin in the DMSCs treatment group was recovered significantly at 4 and 8 weeks after transplantation; the fibroblasts increased at 4 weeks and they distributed at each layer of skin at 8 weeks. There was statistically significant differences between two groups ( P 〈0.01 ). Conclusion Transplantation of DMSCs has an obvious therapeutical effect on the skin photoage.
