摘  要:采用HPLC和GDAS分析测定G-CSF基因工程菌在发酵罐高密度发酵培养过程中菌体产生积累乙酸的含量,分析确定了乙酸含量与产物表达的量化关系,验证了乙酸抑制产物表达的作用。建立了快速测定菌体培养过程中乙酸积累含量检测和动态菌体产物表达量测定的方法。The accumulated content of acetic acid was analyzed by HPLC and the expressed product was determinated by GDAS (Gel Document Analysis System) in the process of high density ferment culture of the G- CSF engineering bacterium. The quantitative relation between the content of acetic acid and the productivity of expressed proteins was established and the restrained effect of acetic acid on the productivity of expressed proteins was proved form the relation. A rapid method in determining the accumulated content of acetic acid and the productivity from the engineering bacterium of the ferment culture was established.
