大足鼠耳蝠MHCⅡ-DRB基因第二外显子的克隆及序列分析

摘  要:为了研究大足鼠耳蝠的MHCⅡ-DRB基因第二外显子的多态性以及大足鼠耳蝠与其他物种的亲缘关系,试验采用PCR产物直接测序方法获得江西和云南两地的大足鼠耳蝠的MHCⅡ-DRB基因第二外显子序列,然后利用BioEdit、Clustal X和Mega软件对测得的序列进行分析,并将其与GenBank中的囊翼蝠、反刍类、灵长类、啮齿类等物种的MHCⅡ-DRB基因第二外显子构建系统发生树。结果表明:大足鼠耳蝠的MHCⅡ-DRB第二外显子序列大小为224 bp,编码74个氨基酸,均具有高度的多态性;江西和云南两地的大足鼠耳蝠聚集为一支,而与包括囊翼蝠在内的其他物种分离开。To research polymorphism of second exon in MHC Ⅱ- DRB gene ofMyotis ricketti and Evolutionary relation among Myotis ricketti and some other species, the sequences of second exon in MHC Ⅱ- DRB gene of Myotis ricketti which from Jiangxi and Yunnan were amplified with direct sequencing of PCR products. Then all these sequences were analyzed by the BioEdit software, Clustal X and Mega. The Phylogenetic relationships among sequences of the second exon of MHC class Ⅱ DRB gene in Myotis ricketti and some other mammals published on GenBank ( including Saccopteryx bilineata, Ruminant, Primate, Rodents, and so on) were constructed by neighbor- joining method. The sequencing data indicated that the full lengths of second exon in MHC Ⅱ - DRB gene of Myotis ricketti were 224 bp ,which encoded 74 amino acids. All of them had Highly polymorphism. The phylogenetic relationships between the Myotis ricketti in JiangXi and YunNan were so closely that could be classified to other mammals including Saccopteryx bilineata.
