摘  要:为研究家鼠型(SEO)汉坦病毒(HV)黑龙江分离株的分子特征,本研究对黑龙江省SEO型新分离株SC106的S基因进行了扩增和序列分析。结果表明:SC106株S基因全长由1775nt组成,编码的蛋白长429aa,符合SEO型编码。与HV参考毒株进行比较,SC106与SEO型同源性最高,与姬鼠型(HTN)相对较低,与其它型别同源性更低。N蛋白进化树分析表明,SC106位于SEO型病毒所在支系,与河南株R22,黑龙江株Pf26和北京株BjHD01亲缘关系接近,体现了一定的宿主依赖性和地理簇集性。序列分析表明:SC106与黑龙江省首个SEO型分离株Pf26均为SEO型中的S1亚型,它们的S基因核苷酸序列同源性也最高(99.2%),并且SC106的S基因编码的N蛋白氨基酸序列也比较保守,由此证实,HV的进化与时间关系不大。The Seoul (SEO) hantavirus SC106 isolated in Heilongjiang area, the S gene of the SC106 was amplified by PCR, sequenced and analyzed. The complete nucleotide sequence of the S gene was composed of 1 775 AT-rich nucleotides which contained one open reading frame encoding for the N protein of 429 amino acid residues. Sequence comparison of the S genes between SC106 and reference hantavirus strains showed that SC106 had higher sequence identities with Hantaan (HTN) viruses than SEO hantavirus and other hantaviruse. Phylogenetic analysis of N proteins showed that SC106 was clustered into the group of SEO type viruses and was more closely related to Henan R22 strain, Heilongjiang Pf26 strain and Beijing BjHD01 strain. The results reflected host-dependent characteristic and geographical aggregation characteristic of hantanviruse. Furthermore, the S gene of Heilongjiang SC106 isolate showed 99.2 % identities with that of the Pf26 strain which was isolated seven years earlier, and the N protein was also well conserved, indicating that the evolution of hantavirus was highly conserved over time.
