摘 要:利用双向凝胶电泳(2-DE)和质谱技术,采用不同剂量60Coγ射线照射人正常肝细胞系HL-7702细胞,观察比较受照细胞克隆子代2-DE图谱的变化,并对差异蛋白质进行鉴定。结果表明,受照肝细胞克隆子代蛋白质表达发生了改变,串联质谱测序和串联质谱数据的Mascot共鉴定出17个差异表达的蛋白质点,4个是上调蛋白质,13个是下调蛋白质;上调蛋白质中,线粒体热休克蛋白60(HSP60)和珠蛋白转录因子1(GATA-1)明显高表达;免疫印迹技术进一步证实HSP60和GATA-1的表达随照射剂量的增加而增强。结果提示,在辐射诱发基因组不稳定性过程中,HSP60和GATA-1蛋白质可能发挥着作用。In this paper the different protein expressions in progeny of irradiated HL 7702 cell line were analyzed by use of two dimensional electrophoresis(2 DE) and matrix assistant laser desorption ion top flight mass spectrometry(MALDI TOF MS).The progeny of irradiated cells were derived from a human liver cell line exposed to 0,2,4,or 6 Gy 60Co rays.Total protein of the cells was extracted using 2 DE and analyzed with ImageMaster 2D Platinum software.In total,42 differentially expressed proteins from the progeny of irradiated cells were screened,of which 17 were identified using MALDI TOF MS analysis.There were 4 up-regulated and 13 down-regulated proteins detected.The up-regulated expression upon ionizing radiation of two proteins,mitochondrial heat shock 60 kD protein(HSP60) and globin transcription factor 1(GATA 1),were further confirmed with immunoblotting.These data suggest that the two proteins may play an important role in initiation and perpetuation of genetic instability in progeny of irradiated human liver cells.
