摘 要:目的观察长波紫外线(UVA)照射所致人真皮成纤维细胞损伤及黄芪甲苷预处理对细胞的保护作用。方法用20μg/ml黄芪甲苷预处理培养人成纤维细胞,2h后以5、10J/cm2剂量的UVA照射细胞并继续培养24h,使用四甲基偶氮唑蓝还原(MTT)法检测细胞活性,以酶联免疫吸附试验(ELISA)检测IL-6和TNF-α的分泌量,比色法检测细胞上清液中的谷胱甘肽过氧化物酶(GSH-Px)和过氧化氢酶(CAT)含量的变化。结果黄芪甲苷对培养的成纤维细胞无明显毒性;UVA照射24h后,成纤维细胞出现增殖活性下降,IL-6和TNF-α分泌量增加,GSH-Px和CAT活力明显下降,黄芪甲苷处理则可抑制上述改变(均P<0.05)。结论黄芪甲苷预处理可明显抑制UVA照射引起的成纤维细胞活性下降,其机制可能与其有效抑制炎性细胞因子分泌和增强细胞抗氧化能力有关。Objective To investigate the protective effects of astragaloside on ultraviolet-A(UVA) induced human fibroblasts damage and its possible mechanisms.Methods Subconfluent fibroblasts were pre-treated with 20 μg/ml astragaloside for 2 h.Then cells were shammed or irradiated with 5,10 J/cm2 UVA irradiation.Cellular activity was detected by MTT assay.The secretion amount of IL-6 and TNF-α was measured by ELISA.And the level of glutathione peroxidase(GSH-Px) and catalase(CAT) were detected by colormetric method.Results The proliferation activity of fibroblasts and the content of GSH-Px and CAT descended with the UVA irradiation,and the secretion of IL-6 and TNF-α was ascensive(P 0.05).But under the intervention of astragaloside,the above changes were inhibited(P 0.05).Conclusion Atragaloside has photo-protective efficiency through strengthening antioxidative efficacy and decreasing cytokine secretion.
