摘  要:目的:探讨血小板活化和哮喘发病的关系。方法:用流式细胞仪对哮喘患者吸入皮质激素治疗前后血小板GMP-140进行测试。结果:哮喘患者治疗前血小板GMP-140表达明显增高(P<0.05);吸入激素治疗6个月后与治疗前有显著差异(P<0.05);治疗后GMP-140正常人无显著差异(P>0.05)。结论:GMP-140可作为血小板活化的特异性指标;GMP-140通过介导血小板粘附聚集参与血小板活化反应,从而直接和/或间接地参与了哮喘发病。Objective:The granule membrane protein-140(GMP-140)on platelets membrane can be acted as a new specific molecular marker of platelet activation.To implore the platelet activation in patients with asthma in remittent stage and the effect of beclomethasone dipropionate inhaler aerosol(bdp).Methods:33 patients with asthma diagnosed according to the criteria of Respiratory branch of the Chinese Medical Association were detected in remittent stages before and after therapy with BDP for 6 months.17 healthy adult volunteer as normal controls without respiratory diseases and other diseases see as reference.Flow cytometric analysis were obtained with gain settings in the logarithmic mode.Results:GMP-140 in asthmatic patients in remission before treatment was significantly increased than those of normal subjects (P<0.05).GMP-140 of asthmatic patients after therapy was significantly lower than those before therapy (P<0.05) and was no obvious difference with normal subjects(P>0.05).FEV 1 value of patients after therapy was higher than those before therapy (P<0.01).Conclusion:GMP-140 is an integrad membrane glycoprotein found in a-granules of human platelets and weibel-palade bodies of human endothelial cells.GMP-140 can induce the adhesion of activated platelet and with blood cells or endothelium in the presence of Ca 2+ ,GMP-140 exists in platelet aggregation in both early and later stages.So we suppose that GMP-140 might take part in the process of asthma,which release inflammatory mediator and oxygen free radical through inducing platelet aggregation.It can be demonstrated that platelet activation exist in asthmatic patients in remission.The mechanism may be related to platelet activation such factors as injured bronchial epithelium inflammatory mediator and inflammatory cells.After therapy there was no statistically significance between patient and normal subjects.We think it is the effect of Corticosteroids.Corticosteroids are not only the most effective antiinflammatory medicine,but also induce the synthesis lipro
