厄贝沙坦对慢性心衰模型鼠B-型利钠肽、细胞因子及心肌细胞凋亡的影响

摘　　要：目的观察厄贝沙坦对心衰模型鼠B-型利钠肽、细胞因子及心肌细胞凋亡的变化。方法将37只SD大鼠随机分为对照组(n=12)和造模组(n=25),造模采用盐酸多柔比星诱导心力衰竭,造模成功随机分模型组和厄贝沙坦(6.75mg/Kg,1次/日)组各12只。厄贝沙坦应用4周后,观察大鼠的一般状况、死亡数、心肌组织形态学的改变、心肌细胞凋亡及细胞凋亡指数,检测B-型利钠肽(BNP)、肿瘤坏死因子-a(TNF-a)及白介素-6(IL-6)水平的变化。结果与对照组相比,模型组大鼠死亡数增加,心肌细胞凋亡指数明显增加,BNP、TNF-a及IL-6水平明显升高,差异显著。与模型组比,厄贝沙坦组大鼠一般状况改善明显,死亡数减少、心肌组织形态学明显改善、心肌细胞凋亡减少及细胞凋亡指数,BNP,TNF-a及IL-6水平明显降低,差异显著。结论模型组大鼠BNP,TNF-a,IL-6水平明显升高,心肌细胞凋亡增加;厄贝沙坦通过降低细胞因子水平,抑制细胞凋亡,从而延缓心力衰竭的进一步发展。Objective To observe effects of Irbesartan on the level of B-type natriuretic peptide,cytokine and cadiocyte apoptosis in the model rats of heart failure.Methods Heart failure induced by doxorubicin were randomly divided into model group（n=12） and Irbesartan group（6.75mg/Kg,once/daily,n=12）,another normal rats were served as control group（n=12）.After four-week treatment,general status,the number of mortality,damaging changes in histomorphology of cardiac muscle in rats and myocardial cell apoptosis were observed;The level of BNP and cytokine were measured.Results In model group,the number of mortality and the index of myocardial cell apoptosis were increased.The level of BNP,TNF-a and IL-6 were remarkably advanced.Irbesartan obviously improved the general status,and decreased the number of mortality;Myocardial cell apoptosis index in Irbesartan group was remarkable lower than model group;There were significant differences in level of BNP,TNF-a and IL-6 between Irbesartan group and model group.Conclusion Irbesartan improved the rats in CHF general status and decreased the number of mortality.It reduced the pathologic damage in myocardium and inhibit myocardial apoptosis,and reduced the level of TNF-a and IL-6 also;Therefore,it maybe delay the progress of heart failure.
