摘 要:目的:研究并建立一种分光光度法用于丹参总酚含量的测定。方法:依据丹参总酚提取物供试品和对照品溶液在紫外或可见扫描图谱中的特征吸收峰一致性,对三氯化铁—铁氰化钾显色法、亚硝酸钠—硝酸铝配位显色法和紫外分光光度法测定丹参总酚含量的可行性进行了比较分析;通过对亚硝酸钠—硝酸铝配位显色法的方法学验证及该法与高效液相色谱法测定的提取物中丹酚酸B的含量的相关性考察,对该法测定丹参总酚酸提取物含量的可行性进行了评价。结果:亚硝酸钠—硝酸铝配位显色法更适于丹参总酚含量的测定,方法学验证的结果表明该法可行,其测定结果与高效液相色谱法测定的结果有明显的相关性。结论:亚硝酸钠—硝酸铝配位显色法可用于丹参总酚的含量测定。Objective:To investigate and establish a spectrophotometry method for determining content of salvianolic acids in the extraction of Salvia miltiorrhiza.Methods:Based on the consistency of the characteristic adsorption bands in the ultraviolet or visible absorption spectum between the reference substance solution and sample solution of SC ,The feasibility of Fe (Cl)3-K3 [Fe (CN)6] colorimetric method,NaNO2-Al (NO3)3 colorimetrie method and ultraviolet spectrophotometry method for determining content of salvianolie acids (SC) was compared and analyzed;The NaNO2-Al(NO3)3 colorimetfic method were studied and evaluated according to the method validation results, and the correlation between the content of SC with spectrophotometry method and Salvianolie acid B with High Performance Liquid Chromatography method.Results:The NaNO2-AI (NO3)3 colorimetric method were more preferable for determining content of SC. the method validation results displayed the method was feasible to determine content of SC,and its determination results had a good correlation with content of Salvianolic acid B with HPLC method.Conclusions:The NaNOE-Al (NO3)3 colorimetric method is feasible for determining content of SC with Salvianolic acid B as the control.
