摘 要:目的观察丙丁酚对高脂高胆固醇饲养的载脂蛋白E(ApoE)基因敲除小鼠和C57BL/6J小鼠肝脏SR-BI和PPARγ表达的影响。方法以载脂蛋白E基因敲除小鼠和具有相同遗传背景的C57BL/6J小鼠为研究对象,饲以高脂高胆固醇饲料和/或丙丁酚。动物喂养12周,摘眼球取血,采用酶比色法及直接检测试剂盒分别测定小鼠血清甘油三酯、总胆固醇、高密度脂蛋白胆固醇和低密度脂蛋白胆固醇的含量;采用HE染色检测肝脏蓄脂情况;免疫组织化学法检测小鼠肝脏SR-BI及PPARγ蛋白质表达;RT-PCR检测小鼠肝脏SR-BI和PPARγ基因的表达。结果血脂检测显示,丙丁酚可以显著降低高脂高胆固醇饲喂的小鼠血清中的TG、TC和LDLC水平(P<0.05)。HE染色显示,丙丁酚处理组小鼠的肝脏脂肪空泡少于未加丙丁酚组。免疫组织化学显示丙丁酚可以上调高脂高胆固醇饲养的小鼠肝脏SR-BI和PPARγ蛋白质的表达(P<0.05)。RT-PCR检测也显示,丙丁酚可以明显增加高脂高胆固醇饲养的C57BL/6J小鼠及ApoE-/-小鼠肝脏SR-BI和PPARγmRNA的表达(P<0.05)。结论丙丁酚可以减轻高脂高胆固醇饲养ApoE-/-小鼠的肝脏蓄脂,其机制可能是上调高脂高胆固醇饲养小鼠肝脏的SR-BI和PPARγ的表达。Objective To investigate the effects of probucol on expressions of scavenger receptor class B type I(SR-BI) and peroxisome proliferators-activated receptor gamma(PPARγ) in ApoE knockout(ApoE-/-) mice and C57BL/6J mice. Methods Twenty male C57 mice and twenty ApoE-/-mice were randomly divided into two groups(n=10,each group),respectively.All of the mice were fed a normal diet enriched with 15% lard and 0.25% cholesterol.The intervention groups were supplemented with 0.5% probucol in HFHC diet.After 12 weeks,all animals were killed.Serum lipids were determined by commercially provided kits.The paraffin embedded sections of thoracic arteries and livers were stained with hematoxylin eosin.Immunohistochemistry assay and reverse transcription-polymerase chain reaction was used to determine the expressions of SR-BI and PPARγ.Results Serum levels of lipids were markedly lowered in mice supplemented with probucol.Probucol decreased the lipid content and upregulated the expressions of SR-BI and PPARγ in the mouse livers,both C57 and ApoE-/-mice.Conclusions Probucol decreases the levels of serum lipids in mice fed with HFHC diet.The lipid lowering and antiatherosclerotic effections may be associated with the upregulation of SR-BI and PPARγ expression in livers.
