摘  要:目的建立一种更实用、标准、可靠的急性大鼠脊髓撞击损伤模型,为脊髓损伤(spinal cord injury,SCI)的进一步研究奠定基础。方法将36只成年雌性Wistar大鼠随机分成三组,每组12只。脊髓损伤(SCI)组:用自制改良的Allen’s撞击器,以60gcm致伤力损伤大鼠T10胸椎对应脊髓。假手术组:只打开椎板,暴露脊髓,不造成SCI。正常对照组:正常大鼠,不做任何处理。各组定期行为学观察(BBB评分),术后30天进行组织学观察和神经电生理检测。结果 HE染色:假手术组与正常对照组基本一致的。SCI组可见灰、白质组织结构不完整,损伤区可见大片坏死灶、细胞肿胀。BBB评分:假手术组术后1周功能恢复接近正常。SCI组术后第2周开始恢复,到第3周基本停止,最终BBB评分未超过6分,两组比较有明显差异。神经电生理(SEP,MEP)检测:SCI组可以明显看到SEP与MEP的峰-峰值急剧降低,且潜伏期明显延长,差异显著(P<0.01)。结论改良后的急性大鼠脊髓损伤模型制作法操作简便、重复性好,是较为理想的方法。Objective To establish a more practical, sandard and dependable model for rats acute spinal cord injury and to lay the foundation of further research on spinal cord injury(SCI). Methods Thirty-six female rats were randomly assigned to 3 groups (n = 12 per group). SCI group: SCI was created with a modified impinger of Allen's by a 60gcm impacting on the T10 spinal cord. Sham group: neural scute was opened only and spinal cord was exposed without SCI. Normal control group:normal rats. Ethological observation (BBB scores) was done regularly. Histological and electmphysiological tests were done on the 30th day after operation. Results HE staining results in Sham group and Normal control group were identical,but SCI group showed fragmented construction in grey matter and necrosis, and cellular swelling was detected in injm^t region. BBB scores revealed that nearly full functional recovery after operation in Sham group required 1Week. In SCI group functional recovery began in 2 nd week after operation and stop in 3 rd week, and the scores failed to exceed 6 in the final. Significant diffe^ence lay between Sham group and SCI group( P 〈 0.01). Electro-physiological(SEP, MEP)demonstrated that peak-to-peak value dropped sharply and latency was extended obviously in SCI group. Conclusion This method can be regarded as a simpler and ideal way which was in good reproducibility to establish the rats model for acute SCI.
