摘 要:目的探讨氟中毒大鼠激活的成骨细胞系中bFGF、c-fos、c-jun的差异表达。方法应用饮水加入氟化钠进行大鼠染氟实验,采用原位杂交技术、Western blot技术、免疫组织化学方法,对氟中毒大鼠骨组织进行增殖刺激因子bFGF、c-fos、c-jun的差异表达的定量分析。结果实验表明无论是在高钙饲养还是在低钙饲养,过量摄入NaF可刺激慢性氟中毒大鼠椎骨各包被成骨细胞增殖,并诱导bFGF过表达,与对照组相比差异显著(P<0.001);c-fos、c-jun表达明显高于对照组,差异显著(P<0.001、P<0.05);相关分析显示bFGF过表达与c-fos、c-jun的蛋白质水平呈正相关。结论过量氟可以刺激大鼠成骨细胞系激活、增殖能力增强、bFGF、c-fos、c-jun表达增强,提示bFGF可能通过上调c-fos、c-jun的表达而参与成骨细胞的增殖和分化。Objective To study the difference expression of bFGF, c-fos, c-jun in activation osteoblasts of fluorosis rat. Methods Rat flurosis model was made by driking water with fluroride sodiuin (NaF), the difference expression of bFGF, c-los, e-jun in acti- vation osteoblasts were analyzed quantitatively by hybridization in situ, Western blot and immmlohistochemistry methods. Results NaF could stimulate the proliferation of periosteal envelop osteoblast in rat with flurosis whether high or low caicium group, induced the over expression of bFGF and were obvious statistic significance compared with control group ( P 〈 0. 001 ). The expression of c-fos/c-jun were higher and obvious statistic significance compared with control group( P 〈 0. 001, P 〈 0.05). The analysis show that the expres- sion of bFGF is rational correlation with the protein expression of c-los, c-jun. Conclusion Excessive fluoride could stimulate the ac- tivation of rat osteoblast , increase the proliferation and increase the expression of bFGF, c-fos, c-jun. It suggested that bFGF maybe take part in the proliferation differation through increasing the expression of c-los, c-jun.
