摘 要:目的观察慢性不可预见性应激(CUS)抑郁模型大鼠行为学改变及血清脑源性神经营养因子(BDNF)蛋白水平的变化,探讨BDNF蛋白在抑郁症发生、发展中的作用。方法 40只大鼠随机分为生理盐水组、氯丙咪嗪组、万拉法新组和正常对照组,每组10只。前3组接受21dCUS制备抑郁大鼠模型,并给予相应的干预。以旷场试验和体质量测定评价药物的疗效,采用酶联免疫吸附试验测定各组大鼠血清BDNF蛋白水平。结果应激后,生理盐水组、氯丙咪嗪组和万拉法新组大鼠旷场试验得分、体质量均低于正常对照组,差别有统计学意义(P<0.05);药物干预后,氯丙咪嗪组、万拉法新组大鼠旷场试验得分、体质量与生理盐水组比较差别有统计学意义(P<0.05);药物干预结束后,氯丙咪嗪组、万拉法辛组和正常对照组大鼠血清BDNF蛋白水平明显高于生理盐水组(P<0.05)。结论 CUS抑郁模型大鼠的行为学表现与临床抑郁症患者精神运动性迟滞极为相似;BDNF蛋白可能在抑郁症的发生、发展过程中起重要作用。Objective To observe the changes of behaviour and brain-derived neurotrophic facotor(BDNF) protein levels in chronic unpredictable stress ( CUS) depression model of rats,and explore the role of BDNF in the occurrence and progression of depression. Methods Forty SD rats were randomly divided into physiological saline group,chlorimipramine group,venlafaxine group and normal control group,ten rats in each group. Chronic unpredictable stress was used to establish the model in physiological saline group,chlorimipramine group and venlafaxine group,the normal control group without any stimulation. The model groups were given corresponding therapy. Open field test were taken to evaluate the curative effect and the enzyme-linked immuno-sorbent assay( ELISA) were taken to detect the BDNF protein levels. Results The open field test scores and body mass in physiological saline group,chlorimipramine group and venlafaxine group were significantly lower than those in the normal control group after stress ( P 〈0. 05) . After treatment,there were statistic differences of the open field test scores and body mass beween chlorimipramine group,venlafaxine group and the physiological saline group ( P〈 0. 05) . The blood serum level of BDNF in psychological saline group was lower than that in the other three groups( P 〈0. 05) . Conclusion The behaviour of depression rat model is very similar to the spirit of sports block of the clinical depression patients,and the BDNF protein may play an important role in the occurrence and procession of depression.
