摘 要:为探讨脱氢表雄酮(DHEA)对肉种鸡脂类代谢的效应机制,选用100羽16周龄AA肉种鸡,随机分成对照组、低剂量试验组、中剂量试验组和高剂量试验组,每组25只。于34周龄开始在饲料中添加DHEA,剂量分别为0 mg/kg、25 mg/kg、50 mg/kg和100 mg/kg,连续饲喂7周。在试验后49 d禁饲12 h,宰杀各组鸡,采集血液和肝脏,测定血清葡萄糖(BG)、总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白-胆固醇(HDL-C)、低密度脂蛋白-胆固醇(LDL-C)和非酯化脂肪酸(NEFA)含量。用放射免疫分析法(RIA)测定血清中三碘甲腺原氨酸(T3)、甲状腺素(T4)、胰高血糖素(Glucagon)、瘦素(Leptin)和肿瘤坏死因子-α(TNF-α)的含量。检测肝脏脂类代谢关键因子乙酰辅酶A羧化酶(ACC)、脂肪酸合成酶(FAS)、肉碱棕榈酰转移酶-Ⅰ(CPT-Ⅰ)和羟甲基戊二酰辅酶A还原酶(HMGR)mRNA水平。结果表明,与对照组相比,中剂量组BG含量显著降低(P<0.05),高剂量组极显著降低(P<0.01);低剂量组和高剂量组HDL-C含量均没有显著差异(P>0.05),中剂量组显著升高(P<0.05);低剂量组和高剂量组NEFA含量均极显著降低(P<0.01),中剂量组差异不显著(P>0.05);各试验组TC、TG和LDL-C含量差异不显著(P>0.05);低剂量组和高剂量组血清T3、T4含量没有明显差异,中剂量组显著升高(P<0.05);低剂量组血清Leptin含量有升高趋势,中剂量组极显著升高(P<0.01),高剂量组显著升高(P<0.05);各实验组Gluca-gon和TNF-α含量均没有显著差异(P>0.05);各试验组肝脏ACCmRNA、FASmRNA水平均呈下降趋势,其中中剂量组显著降低(P<0.05);低剂量组和中剂量组CPT-ⅠmRNA水平均极显著升高(P<0.01),高剂量组显著升高(P<0.05);肝脏HMGRmRNA水平各剂量组均显著下降(P<0.05)。提示,DHEA可能通过提高AA肉种鸡体内脂类分解代谢激素水平及调节肝脏脂类代谢相关基因的表达,从而加快脂肪分解代谢,减少脂肪沉积。In order to study the effect of dehydroepiandrosterone(DHEA) on lipid metabolism of the offspring of AA broilers,one hundred 16-week-old AA broiler breeder were randomly divided into four groups.When the broiler got 34 weeks old,DHEA were added in the feed for 7 weeks at 0 mg/kg,25 mg/kg,50 mg/kg and 100 mg/kg for control group,low-dose DHEA group,medium-dose group and high-dose broils group,respectively.On the 49th day of the experiment,after 12 h fasting,all the breeders were killed.The contents of blood glucose(BG),total cholesterol(TC),triacylglycerol(TG),high density lipoprotein-cholesterol(HDL-C),low density lipoprotein-cholesterol(LDL-C),non-esterified fatty acid(NEFA) in the blood of broilers were detected.The contents of triiodothyronine(T3),thyroxin(T4),glucagon,and leptin and tumor necrosis factor-α(TNF-α) in the blood of broilers were measured by radioimmunoassay(RIA).The mRNA expression of acetyl CoA carboxylase(ACC),fatty acid synthase(FAS),carnitine palmitoyl transferaseⅠ(CPT-Ⅰ) and hydroxy methylglutaryl-CoA reductase(HMGR) were analyzed by real-time PCR.Compared with the control group,the contents of BG droped significantly in high-and medium-dose DHEA group(P0.01 and P0.05,respectively).The content of HDL-C was not affected in low-and high-dose DHEA groups(P0.05),while it was significantly elevated in medium-dose DHEA group(P0.05).The content of NEFA was significantly decreased in low-and high-dose DHEA groups(P0.01).The contents of TC,TG and LDL-C were not influenced in all DHEA treated groups(P0.05).The contents of T3 and T4 did not show big changes in low-and high-dose DHEA groups(P0.05),while they were significantly increased in medium-dose DHEA group(P0.05).The content of leptin kept stable in low-dose DHEA group(P0.05),while it was significantly enhanced in medium-and high-dose DHEA groups(P0.01 and P0.05,respectively).The contents of glucagon and TNF-α remained steady in all DHEA treated
