摘  要:目的:探讨N-乙酰基-丝氨酰-天门冬酰-赖氨酰-脯氨酸(AcSDKP)对转化生长因子-β1(TGF-β1)诱导c-Jun氨基末端激酶(JNK)信号转导通路活化的调节作用.方法:培养新生大鼠肺成纤维细胞.激光扫描共聚焦显微镜观察phospho-JNK在细胞内定位与分布.免疫印迹法检测JNK/phospho-JNK蛋白的表达.结果:激光扫描共聚焦显微镜显示,与对照组比较,TGF-β1刺激后,胞质中phospho-JNK荧光强度变弱,而核浆比值明显增加.经AcSDKP干预作用后,胞质中phospho-JNK的荧光强度较TGF-β1刺激组增强,核浆比值明显减小.免疫印迹法显示,与对照组比较,TGF-β1刺激组phospho-JNK表达明显增加;而AcSDKP干预作用后,phospho-JNK蛋白表达较TGF-β1刺激组明显减少.结论: AcSDKP能阻断TGF-β1介导的肺成纤维细胞JNK通路的活化作用.Objective: To investigate the distribution of phosphorylated JNK protein and the effect of N-acetyl-seryl-aspartyllysyl-proline (AcSDKP) on regulating JNK signal transduction activation mediated by transformig growth factor beta-1 (TGF-β1) in pulmonary fibroblasts. Methods: Cultured neonatal rat pulmonary fibroblasts were used in the experiment. The localization and distribution of phosphorylated JNK was detected by confocal laser scanning microscopy. The expressions of JNK protein and phosphorylated JNK protein in pulmonary fibroblasts were measured by Western blotting. Results: The result of confocal laser scanning microscopy displayed that phosphorylated JNK was located more in the cytoplasm in the control group. Compared with the control group, phosphorylated JNK labeled with green fluorescence in the cytoplasm decreased in the TGF-β1 stimulating group. However, the ratio of nuclei to cytoplasm of fluorescence increased in the TGF-β1groups. Compared with the TGF-β1 stimulating group, the fluorescence in cytoplasm increased and the ratio of nuclei to cytoplasm of fluorescence decreased in AcSDKP treating group. Compared with the control group, the expression of phosphorylated JNK increased significantly in the TGF-β1 group. Compared with the TGF-β1 group, the expression of phosphorylated JNK decreased in the AcSDKP group, while the protein levels of JNK did not change. Conclusion: AcSDKP could block activation of JNK pathway mediated by TGF-β1 in pulmonary fibroblasts.
