摘  要:目的: 探讨Ⅰ型星形胶质细胞条件培养液(ACM-1)对H2O2诱导PC12细胞凋亡的抑制作用与硫氧还蛋白(Trx)表达的关系.方法: 采用MTT法检测ACM-1预处理H2O2诱导PC12细胞的活力.流式细胞术,Real time-PCR法及免疫印迹法分别检测细胞凋亡率,Trx-1和Trx-2mRNA的变化及Trx和Bcl-2蛋白水平的变化.结果: 20%浓度的ACM-1能提高H2O2诱导PC12细胞的活力,细胞凋亡率下降,Trx-1的mRNA量升高,Trx和Bcl-2的蛋白量增加.结论: ACM-1对H2O2诱导PC12细胞凋亡的抑制作用可能是通过Trx上调介导的,而Trx的抗凋亡作用和Bcl-2的上调有关.Objective: To study inhibition of the type I astrocytes conditioned medium (ACM-1) on apoptosis of PC12 induced by H2O2 and the expression level of thioredoxin (Trx) protein. Methods: MTT assay was perfonmed to detect the viability of type I astrocytes-conditioned medium on PC12 induced by H2O2. Flow cytometry, real time-PCR and Western blotting were employed to measure apoptosis rate of cells, mRNA of Trx-1 and Try2, and the expression level of Trx protein and Bcl-2 protein respectively. Results: 20% of ACM-1 increased the viability of PC12 induced by H2O2. The apoptotic rate was significantly decreased compared with the control group. The expresstions of Trx-1 mRNA, Trx and Bcl-2 protein were significantly increased compared with the control group. Condusion: The inhibition effect of type I astrocytes conditioned medium on the apoptosis of PC12 induced by H2O2 is mediated, at least in part, by the up-regulation of Trx expresstion. The antiapoptosis of Trx is correlated with the up-regulated expression of Bcl-2 protein.
