RP-HPLC测定三种黄连中的生物碱含量

摘　　要：目的建立反相高效液相色谱法测定黄连中的原小檗碱型生物碱含量的方法;比较云南黄连、北美黄连、四川黄连生物碱含量的差别。方法采用安捷伦C18色谱柱、乙腈-水为流动相、345nm检测盐酸小檗碱、盐酸巴马亭、盐酸药根碱三种生物碱标准品的出峰时间以及三种黄连三种生物碱含量的多少。并对其进行精密度试验、样品稳定性试验、重现性试验、回收率试验。结果云南黄连、北美黄连、四川黄连三种黄连三种生物碱的含量分别为5.84%、1.26%、0.88%;6.15%、1.35%、0.95%和5.78%、1.38%、1.06%。加回收率分别为103.8%、103.8%、117.4%。结论云南黄连、北美黄连、四川黄连生物碱含量的差别没有统计学意义,本方法检测黄连中生物碱含量的方法快速简便,结果准确、可靠,重现性好。Objective To establish a quantitative method for analyzing the concentration of berberine-type alkaloids in three kinds of coptidis rhizoma in a simultaneous manner using RP-HPLC,and find the different quantity of jatrorrhizine,palmatine,berberine in coptidis rhizoma from Yunnan,Sichuan and north America.Methods Angilent C 18,a mixed mobile phase of acetonitrile-water,and UV detection wavelength：345nm was employed to simultaneously determine the concentration of berberine-type alkaloids in three kinds of coptidis rhizoma.Results The average concentration of three berberine-type alkaloids in coptidis rhizoma was measured at 5.84%,1.26%,0.88%; 6.15%,1.35%,0.95%; 5.78%,1.38%,1.06%. The average recovery rates were registered respectively at 103.8%,103.8%,117.4%.Conclusion This method is simple,accurate,reliable,quick and reproducible. It can be used for quantitative analysis of three berberine-type alkaloids in coptidis rhizoma in a simultaneous manner.
