大黄与活血、止血药配伍调控小鼠腹腔巨噬细胞增殖活性的研究

摘　　要：[目的]观察大黄与丹参、复方丹参、白及药物之间的配伍,及其对小鼠腹腔巨噬细胞增殖活性的调节作用。[方法]建立小鼠腹腔巨噬细胞的分离纯化和培养方法。选用噻唑蓝比色分析方法,测定分析4种中药之间的配伍对巨噬细胞活性的调节作用。每种药物选择3种不同浓度和3个连续时间点。[结果]大黄与白及配伍高浓度、中浓度时对巨噬细胞活性有抑制作用(P<0.01);大黄与丹参配伍高浓度、中浓度及低浓度下对腹腔巨噬细胞增殖活性有促进作用(P<0.01或P<0.05)。大黄与复方丹参配伍仅高浓度、中浓度时对其有促进作用。[结论]体外实验结果表明大黄与不同的药物结成药对可以产生显著不同的效果,为深入研究中药配伍用药提供一些实验数据。[Objective] To observe the compatibility of rhubarb with salvia miltiorrhiza,compound salvia miltiorrhiza and Bletilla striata and the regulatory effect of them on the abdominal macrophage proliferation activity.[Methods] The method of isolation,purification of abdominal macrophage was established.Using MTT colorimetric assay（MCA） the regulatory effect of the four compatible drugs on the macrophages activity was measured with three different concentrations and three sequential time points.[Results] Rhubarb integrated with Bletilla striata at high,middle concentration could inhibit the macrophages activity（P0.05）.On the contrast,Rhubarb with salvia miltiorrhiza at high,middle and low concentration could enhance the proliferation of abdominal macrophage.Rhubarb with compound Salvia miltiorrhiza had an enhancing effect only at high or middle concentration（P0.01 or P0.05）.[Conclusion] The experimental results in vitro showed that Rhubarb combined with different Chinese crude drugs,can produce significantly different effect on the proliferation activity of macrophage,thus providing some data for further research of Chinese drugs compatibility.
