摘 要:目的:优化条件实现人睫状神经营养因子(humanciliaryneurotrophicfactor,hCNTF)cDNA在大肠杆菌中的高效表达。方法:CNTF克隆进pBV220后,表达,凝胶过滤纯化,10日龄鸡背根神经节(E10DRGs)测定其生物活性。结果:CNTF在HB101中高效表达,并具有生物活性。结论:SDS-PAGE观察到相对分子质量约为24×103的预期表达产物,优化表达条件后发现其在HB101中的表达状况最好。Objective: To study high expression of the cDNA of human ciliary neurotrophic factor(hCNTF) in E.coli after obtaining the optimum expression conditions. Methods: CNTF cDNA was cloned into pBV220, and the expressed product was purified with gel filtration after it was highly expressed. Results: CNTF was highly expressed in HB101,which had biological activity after purification. Conclusion: The protein obtained was found to have a relative molecular weight of M r=24×10 3 by SDS PAGE observation and could promote growth of E10 DRG cells.
