白术和黄芪种子试管内萌发试验研究

摘　　要：采用不同消毒方式和培养基对白术和黄芪种子试管内萌发进行探究。实验结果表明:白术种子最佳消毒方式为75%酒精消毒30 s再用0.1%HgCl2消毒5 min;白术种子试管内萌发的适宜培养基为1/2 MS。黄芪种子最佳消毒方式为75%酒精消毒20 s再用0.1%HgCl2消毒10～15 min;黄芪种子试管内萌发的适宜培养基为半固体培养基。The effects of different culture media and disinfection methods on in vitro germination of seeds of Atractylodes macrocephala Koidz.and Astragalus membranaceus（Fisch.） Bge.were studied.The results showed that the optimum disinfection method for seeds of Atractylodes macrocephala Koidz.was firstly treated with 75% ethanol for 30 s and then treated with 0.1% HgCl2 for 5 min;the best culture medium for Atractylodes macrocephala Koidz.seeds was 1/2 MS;the optimum disinfection method for seeds of Astragalus membranaceus（Fisch.） Bge.was firstly treated with 75% ethanol for 20 s and then treated with 0.1% HgCl2 for 10 min to 15 min;the best culture medium for seeds of Astragalus membranaceus（Fisch.） Bge.was semisolid medium.
