摘  要:目的探讨高糖高溶血磷脂胆碱环境下内皮细胞与系膜细胞相互作用对细胞外基质(ECM)积聚的影响及血小板活化因子(PAF)在其中的作用。方法人脐静脉内皮细胞(HUVEC)与人肾小球系膜细胞(HMC)共培养和HMC单独培养,将两种培养模型分别分为对照组、甘露醇组、高糖高脂组、PAF受体拮抗剂BN52021组,比较各组细胞上清液人纤维连接蛋白(Fn)、人Ⅳ型胶原(ColⅣ)、PAF含量及HMC中PAF受体(PAF-R)mRNA的表达水平。结果 (1)高糖高脂促进单培养和共培养细胞Fn、Col Ⅳ和PAF含量的升高,且共培养组Fn、ColⅣ和PAF水平均高于单培养组,BN52021可抑制高糖高脂刺激的Fn和Col Ⅳ升高(P均<0.05)。(2)高糖高脂上调HMC中PAF-R mRNA的表达水平(P<0.05)。结论高糖高脂环境下,系膜细胞和内皮细胞存在异常的相互作用,促进Fn、ColⅣ、PAF的产生,高糖高脂刺激的ECM分泌增加可能与PAF有关。Objective To study the effects of PAF on the relation between the endothelial cells and mesangial cells exposed to high glucose and high lysophosphatidylcholine(LPC). Methods The monoculture of mesangial cell versus endothelial and mesangial cell co-culture were established respectively and divided into 4 groups: control, mannitol, high glucose plus LPC and BN52021. The levels of fibronectin (Fn), collagen Ⅳ(Col Ⅳ) and PAF of supernatant and the expression level of PAF receptor(PAF-R) were measured. Results (1)The levels of Fn, Col Ⅳ and PAF were higher in high glucose plus LPC group than in control group in both co culture and mono culture (P〈0.05). Under the condition of high glucose plus LPC, the levels of Fn, Col IV and PAF were higher in co-culture than in mono-culture(P〈0.05). The levels of Fn and Col Ⅳ were lower in BN52021 group than in high glucose plus LPC group(P〈0.05). (2)The expression level of PAFR in mesangial cells in high glucose plus LPC group was higher than in control group in both co-culture and mono-culture (P〈0.05). Conclusions Under high glucose plus LPC conditions, endothelial cells and mesangial cells can abnormally interact with each other to produce more extracellular matrix and PAF, and the increase of extraeellular matrix is associated with PAF.
